Effect of Scalp Acupoint Penetrating Acupuncture on Alzheimers Disease Rats Based on NF-κB/NLRP3/Caspase-1 Pathway
Objective To analyze the intervention effect of scalp acupoint penetrating acupuncture on Alzheimer's disease rats based on the nuclear factor kappa-B(NF-κB)/NOD-like receptor thermal protein domain associated protein 3(NLRP3)/Caspase-1 pathway.Methods Fifty SPF-grade rats were selected,10 rats were used as the blank group,and the remaining 40 rats were used to establish Alzheimer's disease model.Thirty rats successfully modeled were randomly divided into model group,drug control group and experimental group,10 rats in each group.The rats in the blank group and model group were given normal saline by gavage,while those in the drug control group were treated with Memantine Hydrochloride Tablet by gavage.The rats in the experimental group were treated with acupuncture.The pathological tissues,escape latency,number of platform crossing,oxi-dative stress state,inflammatory factor levels,NF-κB,NLRP3 and Caspase-1 mRNA expression as well as the relative expres-sion of NF-KB/NLRP3/Caspase-1 signaling pathway were observed.Results Compared with those of the blank group,the es-cape latency time and target-platform crossing times were increased in the model group,the drug control group and the experi-mental group.The levels of superoxide dismutase(SOD),catalase(CAT)and interleukin-10(IL-10)were decreased,and the expressions of malondialdehyde(MDA),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),NF-κB,NLRP3 and Caspase-1 mRNA were increased(P<0.05).Compared with those of the model group,the escape latency time was decreased in the drug control group and the experimental group,and the number of platform crossing was increased.The levels of SOD,CAT and IL-10 were increased,and the expressions of MDA,IL-6,TNF-α,NF-κB,NLRP3 and Caspase-1 mRNA were de-creased(P<0.05).Compared with those of the drug control group,the escape latency was decreased and the number of platform crossing was increased in the experimental group.The levels of SOD,CAT and IL-10 were increased,and the expressions of MDA,IL-6,TNF-α,NF-κB,NLRP3 and Caspase-1 mRNA were decreased(P<0.05).Conclusion Scalp acupoint pene-trating acupuncture can improve oxidative stress and reduce the expressions of inflammatory factors in rats with Alzheimer's dis-ease,which may be related to NF-KB/NLRP3/Caspase-1 pathway.
