Effect of Bajitian(Morindae Officinalis Radix)on Hippocampal Neurons Injury of Chronic Stress Depression Rats Based on BDNF/CREB Signaling Pathway
Objective To investigate the effects of Bajitian(Morindae Officinalis Radix)on hippocampal neuronal injury and brain-derived neurotrophic factor(BDNF)/cyclic adenosine phosphos effter-element binding protein(CREB)signaling path-way in rats with chronic stress depression.Methods A total of 40 SD rats were randomly selected as normal group(n=10),and the other rats were randomly divided into 3 groups:model group,fluoxetine hydrochloride group(3.17 mg·kg-1)and Bajitian(Morindae Officinalis Radix)group(3.17 g·kg-1).The rats were administered orally for 8 weeks,once a day.Behavioral tests were performed before and after modeling and after the rats were administered.Hippocampal morphological changes were ob-served by hematoxylin-eosin(HE)staining,the expression of hippocampal BDNF protein was detected by immunohistochemistry(IHC),the pathological damage of hippocampal tissue was observed and scored by HE staining,and the levels of BDNF,TrkB and TRKB were detected by Real-time polymerase chain reaction(PCR).The relative expression of CREB mRNA was detected by Western blot.The relative expressions of BDNF,TrkB and CREB protein were detected by Western blot.Results Compared with those of the normal group,the total activity distance of the model group was significantly decreased(P<0.05),the autono-mous swimming time was significantly decreased(P<0.05),and the tail hanging and struggling time was significantly decreased(P<0.05).HE staining showed the hippocampal neuronal structure was damaged and the pathological injury score was obvious-ly decreased(P<0.05),the expression of BDNF was significantly decreased(P<0.05),and the relative expressions of BDNF,TrkB,CREB mRNA and protein were significantly decreased(P<0.05).Compared with those of the model group,the total activ-ity distance of rats in fluoxetine hydrochloride group and Bajitian(Morindae Officinalis Radix)group was significantly increased(P<0.05),the autonomous swimming time was significantly increased(P<0.05),and the tail hanging struggle time was signifi-cantly increased(P<0.05).The results of HE staining showed that the hippocampal neuronal structure was significantly re-stored,and the pathological injury score was increased(P<0.05).The expression of BDNF in immunohistochemical staining was significantly increased(P<0.05),and the relative expressions of BDNF,TrkB,CREB mRNA and protein were significantly in-creased(P<0.05).Conclusion After chronic stress stimulation,Bajitian(Morindae Officinalis Radix)may exert neuroprotective effects on hippocampal neuronal injury and improve depression-like behavior in depressed rats by activating BDNF/CREB signa-ling pathway.
