Basic Research on Pharmacodynamic Substances of Two Extracts of Shengjiang(Ginger)
Objective By comparing the differences in pharmacological effects and chemical composition of Shengjiang(ginger)water extract and ground juice extract,the pharmacodynamic basis of Shengjiang(ginger)was studied and the rational clinical ap-plication of Shengjiang(ginger)was guided.Methods Using the yeast-induced fever rat model,the antipyretic effects of two ex-tracts of Shengjiang(ginger)were compared.The mice model of xylene induced ear swelling was used to compare the anti-in-flammatory effects of water extract and ground juice extract of Shengjiang(ginger).By comparing the scavenging capacity of water extract and ground juice extract of Shengjiang(ginger)on DPPH free radical,the difference of antioxidant effect was compared.The contents of 6-gingerol,8-gingerol and 10-gingerol in water extract and ground juice extract of Shengjiang(ginger)were determined by HPLC.Ultraviolet spectrophotometry was used to determine the contents of total polysaccharides in water extract and ground juice extract of Shengjiang(ginger).BCA protein concentration detection method was used to determine the total pro-tein content in water extract and ground juice extract of Shengjiang(ginger).Results The water extract of Shengjiang(ginger)had no significant improvement on the yeast-induced fever rat model(P>0.05),while the extract of Shengjiang(ginger)ground juice could significantly improve the yeast-induced fever rat model(P<0.05).The water extract of Shengjiang(ginger)could significantly improve the mice model of xylene induced ear swelling(P<0.05),while the extract of Shengjiang(ginger)ground juice had no significant effect(P>0.05).The scavenging rates of vitamin C,Shengjiang(ginger)water extract and ground juice extract on DPPH free radicals were 79.68%,78.36%and 76.52%,respectively.The contents of 6-gingerol,8-gingerol and 10-gingerol in the water extract of Shengjiang(ginger)were 0.2596 mg/g,0.02718 mg/g and 0.02679 mg/g,respectively,and the contents of 6-gingerol,8-gingerol and 10-gingerol in the Shengjiang(ginger)ground juice were 0.1613 mg/g,0.02082 mg/g and 0.03499 mg/g,respectively.The total polysaccharide contents in the water extract and the ground juice extract of Shengjiang(ginger)were 1.790 mg/g and 1.390 mg/g,respectively.The total protein contents of Shengjiang(ginger)water ex-tract and ground juice extract were 9.90 mg/g and 7.840 mg/g,respectively.Conclusion The antipyretic effect of the water ex-tract of Shengjiang(ginger)is not as good as that of the ground juice extract of Shengjiang(ginger),but the anti-inflammatory effect of the water extract of Shengjiang(ginger)is obviously better than that of the extract of the ground juice extract of Shengjiang(ginger).Both the water extract of Shengjiang(ginger)and the ground juice extract of Shengjiang(ginger)have obvi-ous antioxidant effects.Therefore,when Shengjiang(ginger)is used clinically,its ground juice extract should be selected to play its antipyretic role,its water extract can be selected to play its anti-inflammatory role,and both extracts can play their antioxidant role.The contents of 6-gingerol,8-gingerol,total polysaccharide and total protein in the water extract of Shengjiang(ginger)is relatively more than that in the extract of ground juice,while the content of 10-gingerol is relatively less than that in the extract of ground juice.The difference in the contents of chemical components of the two extracts is the material basis for their different clinical applications.
