滋肾疏肝方对围绝经期抑郁模型小鼠的作用及机制

目的 探讨滋肾疏肝方对围绝经期抑郁(PDD)模型小鼠的作用及其机制。方法 采用卵巢切除法和慢性不可预见性刺激建立PDD小鼠模型。48只小鼠随机分为假手术组,模型组,滋肾疏肝方低、中、高浓度组和阳性对照组,每组8只;低、中、高浓度组分别给予0。38、0。76、1。52 g/mL滋肾疏肝方灌胃,阳性对照组给予0。21 g/mL盐酸氟西汀合戊酸雌二醇混悬液灌胃。通过强迫游泳实验、悬尾实验和旷场实验检测PDD小鼠行为学变化;Nissl染色观察PDD小鼠海马神经元的组织学变化;分别采用qRT-PCR及Western blot实验检测各组小鼠海马神经元脑源性神经营养因子(BDNF)、钙调神经磷酸酶(Calcineurin)、cAMP反应元件结合蛋白(CREB)和CREB调节的转录共激活因子1(CRTC1)基因及蛋白表达;ELISA检测各组小鼠海马组织5-羟色胺(5-HT)、多巴胺(DA)和去甲肾上腺素(NE)水平。结果 PDD模型小鼠增加强迫游泳实验(FST)和悬尾实验(TST)的不动时间,损伤小鼠海马神经元,抑制BDNF、Calcineurin、CREB和CRTC1的表达,增强盐诱导激酶2(SIK2)和p-TRTC1的表达,降低5-HT、DA和NE的水平;而滋肾疏肝方可逆转PDD建模的影响,尤其以中、高浓度组效果显著。结论 滋肾疏肝方可通过CRTC1/CREB/BDNF信号通路,增加单胺类神经递质5-HT、DA、NE的表达,从而修复PDD小鼠海马神经元损伤,发挥抗抑郁作用,为滋肾疏肝方在PDD临床治疗中的应用提供了科学依据,为中医药治疗PDD提供新思路。
Mechanism of Zishen Shugan Decoction(滋肾疏肝方)on Perimenopausal Depression Model Mice
Objective To explore the effect of Zishen Shugan Decoction(滋肾疏肝方,ZSD)on perimenopausal depressive dis-order(PDD)and the underlying molecular mechanism.Methods PDD mouse model was performed by ovariectomy and chronic unpredictable mild stress.Forty-eight mice were randomly divided into sham operation group,model group,ZSD low,medium,high concentration groups and positive control group,with 8 mice in each group.The ZSD low,medium,and high concentration groups were given ZSD with the corresponding concentration(0.38 g/mL,0.76 g/mL and 1.52 g/mL)by gavage,while the posi-tive control group was given 0.21 g/mL of fluoxetine hydrochloride and estradiol valerate suspension by gavage.It observed the behavioral changes in PDD mice through forced swimming experiments,tail suspension experiments and open field experiments.Nissl staining was used to observe the histological changes of hippocampal neurons in PDD mice.The qRT-PCR and Western blot experiments were used to detect the expression of brain derived neurotrophic factor(BDNF),calcineurin,cAMP responsive element binding protein(CREB)and transcriptional coactivator 1(CRTC1)regulated by CREB in the hippocampal neurons of mice in each group.ELISA detected the levels of 5-hydroxytryptamine(5-HT),dopamine(DA),and norepinephrine(NE)in the hippocampus of mice in each group.Results PDD model mice increased the immobility time of forced swimming test(FST)and tail suspension test(TST),damaged hippocampal neurons,inhibited the expressions of BDNF,calcineurin,CREB and CRTC1,enhanced the expressions of salt induced kinase 2(SIK2)and p-TRTC1 and reduced the levels of 5-HT,DA and NE.ZSD can reverse the impact of PDD modeling,especially in the ZSD medium and high concentration groups.Conclusion ZSD can increase the expressions of monoamine neurotransmitters 5-HT,DA and NE through the CRTC1/CREB/BDNF signaling path-way,thereby repairing the hippocampal neuronal damage in PDD mice and exerting antidepressant effects.It provides a scientific basis for the application of ZSD in the clinical treatment of PDD and new ideas for the treatment of PDD with traditional Chinese medicine.
