Study on Pugongying(Taraxaci Herba)Material Basis for Clearing Heat and Detoxifying Based on Anti-inflammatory Effect
Objective To explore the pharmacodynamic basis of Pugongying(Taraxaci Herba)for clearing heat and detoxif-ying.Methods The anti-inflammatory effects of Pugongying(Taraxaci Herba)fresh homogenate,fresh water extract and dried Pugongying(Taraxaci Herba)water extract were compared by using xylene-induced ear swelling model.High performance liq-uid chromatography-tandem mass spectrometry(HPLC-MS/MS)method was used to compare the contents of five components including sub-biomass luteolin,chlorogenic acid,ferulic acid,caffeic acid and ethyl pigment in Pugongying(Taraxaci Herba)fresh homogenate,fresh water extract and dried Pugongying(Taraxaci Herba)water extract.The phenol-sulfuric acid colorimet-ric method and Coomassie brilliant blue method were used to compare the differences of primary biomass polysaccharides and pro-tein contents in Pugongying(Taraxaci Herba)fresh homogenate,fresh water extract and dried Pugongying(Taraxaci Herba)water extract,respectively.The polysaccharides and protein components in Pugongying(Taraxaci Herba)fresh products were separated and purified by ion exchange chromatography,and the material basis for clearing heat and detoxification was further studied.Re-sults The ear swelling rates of Pugongying(Taraxaci Herba)fresh homogenate,fresh water extract and dried Pugongying(Taraxaci Herba)water extract group were 0.59,0.85 and 0.94,respectively,among which the content of sub-biomass in Pugongying(Taraxaci Herba)fresh homogenate extract was the lowest,while the contents of primary biomass polysaccharides and protein components were the highest,and the ear swelling rates of mice in the polysaccharide group and protein group were 0.45 and 0.74,respectively.Conclusion The anti-inflammatory effect of Pugongying(Taraxaci Herba)fresh products is better than that of dried products,and the difference between the content of sub-biomass in fresh and dried products is small,and the contents of polysaccharides and proteins in primary biomass are very different.
