基于信号通路探讨桂枝茯苓丸治疗卵巢癌的研究进展

摘要
桂枝茯苓丸是治妇人癥病的中医经典名方,现代研究已经证明其具有抗纤维化、抗炎、抗肿瘤、调节机体免疫、改善脑缺血等作用.卵巢癌(Ovarian Cancer,OC)是女性生殖系统常见的恶性肿瘤之一,早期症状少不易诊断,晚期临床疗效不佳,严重威胁着妇女的健康.通过检索中国知网(CNKI),PubMed,万方(Wanfang Data),维普(VIP)等数据库,从桂枝茯苓丸及其方中单味药主要成分入手,归纳整理了近年来桂枝茯苓丸及其方药主要成分治疗OC的相关通路,发现其主要涉及 PI3K/Akt/mTOR、JAK2/STAT3、Wnt/β-catenin、NF-κB、VEGF/VEGFR2、Notch 多条信号通路.桂枝茯苓丸通过以上多条信号通路发挥抑制肿瘤细胞增殖、促进细胞凋亡、阻滞细胞周期、抗肿瘤血管生成等功效,实现良好的抗肿瘤作用,为临床预防和治疗OC提供强有力的支撑依据,更好地体现出中药复方治疗疾病多途径、多靶点的作用优势.
Abstract
Guizhi Fuling Pill(桂枝茯苓丸),a classic traditional Chinese medicine(TCM)formula,has been verified in modern research to possess various pharmacological effects such as anti-fibrotic,anti-inflammatory,anti-tumor,modulating body immunity and improving cerebral ischemia effects.Ovarian cancer(OC)is one of the common malignant tumors of the fe-male reproductive system and is difficult to diagnose with few early symptoms and poor clinical outcomes in the late stages,posing a serious threat to women's health.By searching from such databases as China National Knowledge Infrastructure(CNKI),PubMed,Wanfang Data Knowledge Service Platform and Chongqing Weipu Database for Chinese Technical Periodicals(VIP),this article summarized the relevant pathways involved in the treatment of OC by Guizhi Fuling Pill and the main ingredients of its for-mula,and has found that they mainly involved phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR),Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3),Wnt/β-catenin,nu-clear factor kappa-B(NF-κB),vascular endothelial growth factor(VEGF)/vascular endothelial growth factor receptor 2(VEG-FR2)and Notch multiple signaling pathways.Guizhi Fuling Pill inhibits tumour cell proliferation,promotes apoptosis,blocks cell cycle and resists tumour angiogenesis through the above-mentioned signaling pathways to achieve good anti-tumour effects,pro-viding a strong basis for clinical prevention and treatment of OC and better reflecting the advantages of multiple pathways and tar-gets in the treatment of diseases by Chinese herbal medicine compound.
