重楼皂苷Ⅰ抗肿瘤作用研究进展

恶性肿瘤已成为威胁全球健康的重大难题之一。化疗和靶向治疗是其重要的药物治疗手段,但由于药物不良反应和耐药性严重影响患者的治疗依从性、生活质量和治疗效果。越来越多的研究表明,中草药是探索新型抗癌疗法的天然宝库,具有很高的医用价值。清热解毒法是中医治疗恶性肿瘤的基本法则之一。清热解毒类中草药重楼,其性微寒,味苦,有小毒,归足厥阴肝经,具有消肿止痛、清热解毒、凉肝定惊之功效。重楼主要适用于疔疮痈肿、咽喉肿痛、蛇虫咬伤、跌打损伤以及惊风抽搐等症。重楼皂苷Ⅰ是中草药重楼的有效活性成分之一,其化学结构为C44H70O16。大量实验表明,重楼皂苷V能通过抑制肿瘤细胞增殖、阻滞细胞周期、诱导细胞凋亡自噬、逆转肿瘤耐药、抑制肿瘤血管生成、抑制肿瘤EMT、调节肿瘤微环境等途径抑制多种恶性肿瘤细胞生长。查阅国内外关于重楼皂苷Ⅰ抗肿瘤作用及机制的相关文献,对其进行归纳总结,为重楼皂苷Ⅰ抗肿瘤作用机制深入研究及抗肿瘤新药研发提供相应参考。
Research Progress on Anti-tumor Effects of Polyphyllin Ⅰ
Cancer is one of the major health challenges threatening the world.Chemotherapy and targeted therapy are impor-tant treatments for malignant tumors,but the severe side effects and drug resistance greatly affect patients'treatment compliance,quality of life and treatment efficacy.A lot of studies have revealed that traditional Chinese medicine is a natural treasure for ex-ploring novel anti-cancer therapies.The traditional Chinese herb Chonglou(Paridis Rhizoma),which belongs to the category of clearing heat and detoxifying herbs,is slightly cold and bitter,and has a mild toxicity.It has diverse medical effects,such as re-ducing swelling and pain,clearing heat and detoxifying.It is applicable for furuncle,sore throat,snake and insect bites,bruises and convulsions.Polyphyllin Ⅰ is the main bioactive component in Chonglou(Paridis Rhizoma).The chemical formula is C44H70 O16.Numerous experiments have shown that Polyphyllin Ⅰ can inhibit the growth of various malignant tumor cells by inhibiting tumor cell proliferation,blocking cell cycle,inducing apoptosis and autophagy,reversing tumor resistance,inhibiting tumor angio-genesis,inhibiting tumor EMT and regulating tumor microenvironment.Based on the relevant literatures of Polyphyllin Ⅰ in Chi-na and abroad,the present study reviewed the research progress of Polyphyllin Ⅰ to provide useful references for the further study of the anti-tumor effect of Polyphyllin Ⅰ and the development of novel anti-tumor drugs.
