Experimental Study on Effect of Triple-needling at "Huantiao(GB30)" on Expressions of Inflammatory Factors and GFAP in Spinal Dorsal Horn of Rats with Sciatic Nerve Injury
Objective To investigate the potential analgesic mechanism of triple-needling at"Huantiao(GB30)"in the treat-ment of neuropathic pain by observing the effect of triple-needling at"Huantiao(GB30)"on inflammatory factors and glial fi-brillary acidic protein(GFAP)expression in the spinal dorsal horn of rats with sciatic nerve injury.Methods Sixty SD rats were randomly divided into three groups:sham operation group,model group,triple-needling group,single needling group and drug group,with 12 rats in each group.The rat sciatic nerve injury model was prepared by clamp method.The acupuncture treatment was started on the second day of modeling,once a day,15 minutes for each time,totally for 14 days.The changes of paw with-drawal latency(PWL)of rats in each group were observed before and after intervention.After treatment,the injured sciatic nerve was taken out and the nerve morphology was observed with HE staining.The expression levels of interleukin-1β(IL-1β),in-terleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)protein were detected by ELISA and the expression level of GFAP was detected by RT-PCR and immunohistochemistry method.Results Before treatment,compared with that of the sham group,the PWL index in each group decreased(P<0.01),and compared with that of the model group,the PWL index in the triple-needling group was significantly improved(P<0.01).HE staining showed that the arrangement of nerve fibers in the model group was disordered,and the number and arrangement of nerve fibers in the triple-needling group,single needling group and drug group were improved,and the improvement in the triple-needling group was the most obvious.ELISA,RT-PCR and im-munofluorescence detection showed that the expressions of IL-1β,IL-6,TNF-α and GFAP in the model group,the triple-needling group,the single needling group and the drug group were significantly higher than those in the sham group(P<0.01).Compared with those of the model group,the expressions of IL-1β,IL-6,TNF-α and GFAP in the triple-needling group,single needling group and drug group decreased significantly,and the expression in the triple-needling group was lower than that in the single needling group and drug group(P<0.01).Conclusion The analgesic mechanism of triple-needling at"Huantiao(GB30)"in relieving neuropathic pain caused by sciatic nerve injury may be related to down-regulating the expressions of in-flammatory factors in the dorsal horn of the spinal cord,inhibiting the activation of astrocytes and reducing the degree of central pain sensitization.
