摘要

目的 优选白术多糖的提取分离工艺.方法 比较氯仿-正丁醇Sevage法去蛋白与不去蛋白白术多糖含量.以吸附率和解吸率为考察指标,优选AB-8型、D101型、LX-38型3种大孔吸附树脂.通过吸附时间、解吸时间、上样流速、上样浓度、洗脱剂用量、洗脱剂浓度和洗脱流速研究AB-8型大孔树脂纯化白术多糖工艺.结果 未去蛋白的多糖含量为(5.582±1.448 0)%、(5.407±0.766 8)%、(5.155±0.721 4)%,Sevage 法去蛋白后的多糖含量为(4.658±0.155 8)%、(4.247±0.074 2)%、(4.610±0.173 4)%.AB-8型大孔树脂较其他两种树脂,其吸附率及解吸率可达(18.40±0.030 0)%,(91.73±0.078 1)%,纯化工艺为吸附时间3 h、解吸时间3 h、上样流速1 BV/h、上样浓度0.75 mg/mL、洗脱剂用量4BV、洗脱剂浓度15%乙醇、洗脱流速3 BV/h.结论 本试验为白术多糖的提取分离提供了试验依据,为白术多糖的工业化生产提供了参考.

Abstract

Objective To optimize the extraction and separation process of polysaccharides from Baizhu(Atractylodes macro-cephala).Method The contents of polysaccharides from Baizhu(Atractylodes macrocephala)without protein removal and protein removal by Sevage method were compared.Three macroporous adsorption resins of AB-8,D101 and LX-38 were investigated based on adsorption rate and desorption rate.The purification process of AB-8 macroporous adsorption resin was studied by ad-sorption time,desorption time,sample flow rate,sample concentration,eluent dosage,eluent concentration and elution flow rate.Result The contents of polysaccharides without removing protein were(5.582±1.448 0)%,(5.407±0.766 8)%and(5.155±0.721 4)%.The contents of polysaccharides after removing protein by Sevage method were(4.658±0.155 8)%,(4.247±0.074 2)%and(4.610±0.173 4)%.The adsorption rate and desorption rate of AB-8 macroporous resin were(18.40±0.030 0)%and(91.73±0.078 1)%respectively.The purification process was as follows:adsorption time 3 h,de-sorption time 3 h,sample flow rate 1 BV/h,sample concentration 0.75 mg/mL,eluent dosage 4 BV,eluent concentration 15%ethanol,elution flow rate 3 BV/h.Conclusion The experiment provided the experimental basis for the extraction and separation of and a reference for the industrial production of polysaccharides from Baizhu(Atractylodes macrocephala).
