N6-甲基腺嘌呤甲基化:中医药防治疾病的新兴靶点

N6-甲基腺嘌呤(N6-methyladenosine,m6A)修饰是真核细胞信使RNA(messenger RNA,mRNA)最常见的一种表观遗传修饰方式,近年来发现其在疾病的发生发展中扮演重要角色。通过国内外权威数据库检索分析,阐述了m6A修饰的概念、作用机制及其研究背景,介绍了参与调控m6A的三大类甲基化酶在m6A修饰过程中扮演的角色及其在疾病发生发展中的作用,并对近年来中药及其有效成分通过干预m6A修饰防治疾病的相关研究成果进行了系统地梳理和归纳发现,中药及其有效成分是宝贵的天然m6A调节剂,其在防治癌症、心血管疾病、生殖系统疾病及骨相关等疾病中具有较好的干预作用,但其干预m6A修饰防治疾病的研究正处于探索阶段,进一步探析其具体网络调控机制无疑是未来中医药研究的热门方向。为今后进一步探索中医药通过m6A修饰治疗疾病的潜在价值,挖掘开发可调控表观遗传的中药提供理论参考及依据。
N6-Methyladenosine Methylation:A New Target for Prevention and Treatment of Diseases by Traditional Chinese Medicine
N6-methyladenosine(m6A)modification is the most common epigenetic modification of eukaryotic messenger RNA(mRNA).In recent years,it has been found that m6A plays an important role in the occurrence and development of disea-ses.Through the retrieval and analysis of authoritative databases at home and abroad,this paper expounds the concept,mecha-nism and research background of m6A modification,and introduces the role of three kinds of methylases involved in regulating m6A modification in the process of m6A modification and their role in the occurrence and development of diseases.In addition,it systematically combs and summarizes the relevant research achievements of traditional Chinese medicine and its active compo-nents in preventing and treating diseases by interfering with m6A modification in recent years.Traditional Chinese medicine and its active ingredients are the valuable natural m6A regulators and have a good intervention effect in the prevention and treatment of cancer,cardiovascular diseases,reproductive system diseases and bone-related diseases,but its intervention in the prevention and treatment of diseases modified by m6A is still in the exploratory stage.Further exploration of its specific network regulation mechanism is undoubtedly a hot direction of traditional Chinese medicine research in the future.This paper can provide theoreti-cal reference and basis for further exploring the potential value of traditional Chinese medicine through m6A modification in the treatment of diseases and excavating and developing traditional Chinese medicine which can regulate epigenetics.
