Contents of 10 Components Differences of Heart-Nourishing and Mind-Easing Drug Pair of Suanzaoren(Ziziphi Spinosae Semen)-Wuweizi(Schisandrae Chinensis Fructus)Before and After Decocting Based on UPLC-Q-TRAP-MS
Objective To analyze the main index constituents differences of the heart-nourishing and mind-easing drug pair of Suanzaoren(Ziziphi Spinosae Semen)-Wuweizi(Schisandrae Chinensis Fructus)before and after decocting and establish a method for content determination.Methods Using Welch Ultimate XB-C18(2.1 mm ×50 mm,5 μm)column,the mobile phase was gradient elution with acetonitrile 0.1％formic acid solution.The flow rate was 0.4 mL/min and the column temperature was 40 ℃.The mass spectrometry was performed by electrospray ion source(ESI)with multi-reaction monitoring(MRM)mode.Jujuboside A,Spinosin,Schisandrin,Schisantherin A,Schisandrin A,Schisandrin B,Rutin,Quercetin,Kaempferol and Kaempferol-3-0-rutinoside were separately determined and the content differences of main index constituents were analyzed before and after compatibility.Results The linear relationship of the above 10 components was good in their respective concentra-tion ranges.The RSD values of precision,repeatability and stability were good.The average recoveries were 95.0％～102.1％,and RSD value was 0.03％～3.7％.The results of this experiment were consistent with those of previous experiment that the main index components of Suanzaoren(Ziziphi Spinosae Semen)-Wuweizi(Schisandrae Chinensis Fructus)decreased after de-coction.Conclusion The simultaneous determination of multiple indicator components provides a new method for comprehensive control of the compatibility quality of this drug and a basis for in-depth research on its pharmacological substance basis.
