Effect of Majing Xuanfei Granules(麻荆宣肺颗粒)on Human Coronavirus 229E Mouse Pneumonia Model
Objective To study the efficacy of Majing Xuanfei Granules(麻荆宣肺颗粒)on human coronavirus 229E infected mouse model and provide reference for its clinical application.Methods The human coronavirus 229E infected mouse model was used in this study.On the day of infection,Majing Xuanfei Keli Granules were administered for four days.On the fifth day,the whole blood was collected from orbit after measuring the weight.The lung tissue was dissected,and the lung index and inhibition rate were calculated.The coronavirus 229E virus load in lung tissue was detected by Real-time PCR.The percentage of immu-nocytes was detected by flow cytometry.The contents of inflammatory factors in lung tissue and serum were detected by ELISA.The levels of angiotensin converting enzyme 2(ACE2)and mitochondrial assembly receptor 1(MAS1)in lung tissue were detec-ted by immunohistochemical method.Histopathological analysis was performed with lung tissue.Results The result showed that Majing Xuanfei Granules significantly reduced the lung index of human coronavirus 229E infected mouse model(P<0.01),and the inhibition rates were 66.80%,53.20%and 45.20%,respectively.Majing Xuanfei Granules also significantly reduced the lung tissue viral load(P<0.01),increased the peripheral blood lymphocyte percentage(P<0.05,P<0.01),reduced the in-flammatory factors levels in lung tissue and serum(P<0.01),increased the expressions of ACE2 and MAS1 in lung tissue(P<0.05,P<0.01)and improved the bronchial and alveolar tissue inflammatory lesions.Conclusion Majing Xuanfei Granules showed a therapeutic effect on human coronavirus 229E infected mouse model.The mechanism may be related to virus replication inhibition,immunological regulation and inflammatory factor reduction.
