Effects of Huangqi(Astragali Radix)on Expression of VDR and Level of IL-23 and IL-17 in Skin Lesions of Psoriatic Mice Models
Objective To observe the effect of Huangqi(Astragali Radix)on the expression of vitamin D receptor(VDR)and the levels of interleukin-23(IL-23)and interleukin-17(IL-17)in psoriatic lesions induced by iniquitous in mice.Methods The successfully established mice were randomly divided into model control group,vitamin D 30 ng/kg group,methotrexate 1 mg/kg group and Huangqi(Astragali Radix)2.4 g/kg,4.8 g/kg and 9.5 g/kg groups.Another normal control group was estab-lished,and the intervention lasted for 3 weeks.The mouse skin lesions were scored using the skin lesion severity index(PASI)score.HE staining was used to observe the histopathology of mouse skin lesions.The serum levels of 25(OH)D3,IL-23 and IL-17 in mice were detected by ELISA.Real-Time qPCR was used to detect the expression of VDR mRNA in mouse skin le-sions.Immunohistochemistry method was used to detect the expression of VDR protein in mouse skin lesions.Results Compared with the normal control group,the model group showed significant changes in skin lesions,with a significant increase in PASI score(P<0.05),characteristic pathological changes in psoriasis,a significant decrease in serum 25(OH)D3 levels(P<0.05),a significant increase in levels of IL-23 and IL-17(P<0.05),and a significant down-regulation of VDR mRNA and protein expression in skin lesions(P<0.05).Compared with the model control group,the PASI scores in the Huangqi(Astragali Radix)4.8 g/kg and 9.5 g/kg,vitamin D and methotrexate groups were significantly reduced(P<0.05).Conclusion Huangqi(Astra-gali Radix)intervention in psoriasis by correcting the imbalance of the levels of IL-23 and IL-17 may be related to regulating the expression of VDR.
