Effect and Mechanism of Yinang Zhuyun Decoction(抑囊助孕汤)on Inflammatory State in Rats with Polycystic Ovary Syndrome
Objective To investigate the effect of Yinang Zhuyun Decoction(抑囊助孕汤)on inhibitor nuclear factor κB-α/nuclear transcription factor-κB(IκB-α/NF-κB)signaling pathway in ovarian tissue of polycystic ovary syndrome(PCOS)rats,in order to explore the further mechanism of Yinang Zhuyun Decoction in the treatment of PCOS from the perspective of in-flammation.Methods A total of 60 female SPF SD rats were randomly divided into blank control group(n=12)and pathological model group(n=48).Pathological model rats were prepared by continuous intragastric administration of letrozole(0.001 g/kg)for 21 days.After successful modeling,the rats in the pathological model group were randomly divided into four groups including model group,Diane-35 group,low dose group combined with Diane-35 and high dose group combined with Diane-35,each group consisted of 12 animals.After 28 d drug intervention,the abdominal aortic blood and bilateral ovaries of rats in each group were taken out.The mass of the ovaries was weighed.Ovary tissue and morphology were observed by hematoxylin-eosin(HE)staining.Enzyme-linked immunosorbent assay(ELISA)was used to detect testosterone,luteinizing hormone(LH)and inflam-matory factors including tumor necrosis factor-α(TNF-α),interleukin-18(IL-18),interleukin-6(IL-6)and C-reactive protein(CRP).Western blotting was used to detect NF-κB p65 and IκB-α protein expressions in ovarian tissue of rats.Results Compared with those of the blank control group,the mass of the ovaries,atretic follicles in the ovaries,the serum levels of testosterone,LH,TNF-α,IL-18,IL-6 and CRP in the model group were significantly increased(P<0.05),the expression of NF-κB p65 protein was increased,and the expression of IκB-α was decreased(P<0.05).Compared with the model group,the Yinang Zhuyun Decoction and Danyin-35 had a significant reduction in ovarian weight(P<0.05),the number of atretic fol-licles decreased and the number of corpus luteum increased.The serum levels of testosterone,LH,TNF-α,IL-18,IL-6 and CRP in the model group were significantly reduced(P<0.05).The expression of NF-κB p65 protein in ovarian tissue had a significant reduction and the protein expression of IκB-α was significantly increased(P<0.05).Conclusion Yinang Zhuyun Decoction could inhibit the inflammatory response of PCOS rats through IκB-α/NF-κB signaling pathway,and diminish the chronic low-grade inflammation state of PCOS rats.
