Heshouwu(Radix Polygoni Multiflori)-Niuxi(Achyranthis Bidentatae Radix)Anti-Vitaminic Acid-Induced Osteoporosis and Screening of Active Ingredients
Objective To study the anti-osteoporosis effect of Heshouwu(Radix Polygoni Multiflori)-Niuxi(Achyranthis Bi-dentatae Radix)and the screening of activity ingredients.Methods HPLC-MS was used to analyze the common fingerprint peaks of Heshouwu(Radix Polygoni Multiflori)-Niuxi(Achyranthis Bidentatae Radix)and confirm their components.The vitaminic acid was used to copy the osteoporosis model and ELISA method was used to explore the anti-osteoporosis effect of Heshouwu(Radix Polygoni Multiflori)-Niuxi(Achyranthis Bidentatae Radix).The correlation of chromatographic results with the wet weights of liver and kidney,the ration of dry weight and wet weight of the bones and ELISA measurement results was analyzed by SPSS for screening the active ingredients of Heshouwu(Radix Polygoni Multiflori)-Niuxi(Achyranthis Bidentatae Radix)anti-osteoporosis.Results A total of 10 common fingerprint peak components were identified by HPLC-MS analysis:gallic acid,cyas-terone,β-ecdysterone,25-R-achyosterone,stilbene glucoside,25-S-achyosterone,emodino,physcion,oleanolic acid 3-O-β-D-glucuro-nopyranoside,achyranthoside D.The results of biological indicators showed that Heshouwu(Radix Polygoni Multiflori)-Niuxi(Achyranthis Bidentatae Radix)can increase the bone alkaline phosphatase(BALP)and bone gla protein(BGP-OCN)indicators,and reduced the levels of tartrate resistant acid phosphatase 5b(TRACP-5b)and C-terminal te-lopeptides of type Ⅰ collagen(CTX-1)of the osteoporosis rats caused by osteic acid.There was significant decrease in liver and kidney indexes,increase in the ratio of dry weight and wet weight of the bones and significant weight change after Heshouwu(Ra-dix Polygoni Multiflori)-Niuxi(Achyranthis Bidentatae Radix)treatment.Conclusion Heshouwu(Radix Polygoni Multiflori)-Niuxi(Achyranthis Bidentatae Radix)has anti-osteoporotic effect,and the most significant active ingredients were gallic acid,β-ecdysterone,emodin and oleanolic acid-3-O-β-D-glucuro-nopyranoside.
