化痰活血通络方调节肠道菌群抑制脂多糖介导的炎症通路改善动脉粥样硬化小鼠血管损伤

目的 从肠道菌群探讨化痰活血通络方治疗动脉粥样硬化(atherosclerosis,AS)的机制,为化痰活血通络方临床应用提供依据。方法 25只雄性ApoE-/-小鼠,高脂饲料喂养12周,制备AS模型;分为模型组、全方组、通络组、化痰组、他汀组各5只,另设C57BL/6J雄性小鼠5只作为正常组,各组相应药物灌胃治疗12周。采用16S rRNA扩增子测序检测肠道菌群;苏木素-伊红(HE)染色法观察小鼠主动脉及肠道的病理形态;酶联免疫吸附测定法(ELISA)检测小鼠血清中相关指标,Western blot法检测Toll样受体4(TLR4)、NOD样受体热蛋白结构域相关蛋白3(NLRP3)、核因子-κB(NF-κB)、白介素-6(IL-6)蛋白含量。结果 与正常组相比,模型组中物种丰富度及多样性下降明显,厚壁菌门(Fir-micutes)与拟杆菌门(Bacteroidetes)的比例(F/B)显著上升,血清中脂多糖(LPS)、甘油三酯(TG)、总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C),主动脉中TLR4、NLRP3、NF-κB、IL-6蛋白含量显著上升(P<0。01),高密度脂蛋白胆固醇(HDL-C)含量显著下降(P<0。01),小鼠主动脉斑块面积显著增加(P<0。01)并伴有大量泡沫细胞及炎性浸润,肠内有大片绒毛脱落与固有层裸露,与模型组相比全方组可显著增加物种丰富度及多样性,降低F/B的比例(P<0。01),提高有益菌的相对丰度,降低有害菌的相对丰度,在全方组与阿托伐他汀组中LPS、TG、TC、LDL-C、TLR4、NLRP3、NF-κB、IL-6均显著下降(P<0。01),HDL-C显著上升(P<0。01),并且可明显缓解上述病理改变。结论 化痰活血通络方可改善肠道菌群的组成及脂代谢,减少LPS的产生,并通过TLR4/NF-KB/NLRP3信号通路下调主动脉中炎性因子的表达,减少脂质的积累来缓解ApoE-/-小鼠的动脉粥样硬化。
Huatan Huoxue Tongluo Recipe(化痰活血通络方)Regulates Intestinal Microflora and Inhibits LPS Mediated Inflammatory Pathway to Improve Vascular Injury in Atherosclerosis Mice
Objective To explore the mechanism of Huatan Huoxue Tongluo Recipe(化痰活血通络方)in treating atheroscle-rosis(AS)from intestinal flora and provide a basis for the clinical application of Huatan Huoxue Tongluo Recipe.Method Twenty-five male ApoE-/-mice were fed high fat diet for 12 weeks to prepare AS models.The model mice were divided into model group(MX),whole recipe group(QF),dredging collateral group(TL),resolving phlegm group(HT)and atorvastatin group(XY)with 5 mice each.In addition,5 C57BL/6J male mice were set as the normal group(ZC),and each group was treated with the corresponding drugs by gavage for 12 weeks.The gut microbiota was detected by using 16S rRNA amplicon sequencing.The pathological morphology of mouse aorta and intestine was observed by using hematoxylin eosin(HE)staining method.Enzyme-linked immunosorbent assay(ELISA)was used to detect relevant indicators in mouse serum.Western blot was used to detect Toll like receptor 4(TLR4),NOD like receptor heat protein domain related protein 3(NLRP3),nuclear factors-κB(NF-κB)and interleukin-6(IL-6)protein contents.Result Compared with those in the blank group,the species richness and diversity in the MX group decreased significantly,and the proportion of Firmicutes to Bacteroidetes(F/B)increased significantly.Lipopo-lysaccharides(LPS),triglycerides(TG),total cholesterol(TC)and low-density lipoprotein cholesterol(LDL-C)in the serum as well as TLR4,NLRP3 and NF-κB in the aorta were observed in the MX group.The content of IL-6 protein increased signif-icantly(P<0.01),the content of high-density lipoprotein cholesterol(HDL-C)decreased significantly(P<0.01),the area of aortic plaque in mice increased significantly(P<0.01),accompanied by a large number of foam cells and inflammatory infiltra-tion,large areas of villus shed and lamina propria exposed in the intestine.Compared with the MX group,the QF group could sig-nificantly increase the species richness and diversity,reduce the proportion of F/B(P<0.01),and increase the relative abun-dance of beneficial bacteria,reduce the relative abundance of harmful bacteria,including LPS,TG,TC,LDL-C,TLR4,NLRP3 and NF-κB in the QF group and XY group.IL-6 significantly decreased(P<0.01),HDL-C significantly increased(P<0.01),and the above pathological changes were significantly alleviated.Conclusion Huatan Huoxue Tongluo Recipe can improve the composition of intestinal microbiota and lipid metabolism and reduce the production of LPS,and it can down regulate the ex-pression of inflammatory factors in the aorta and reduce the accumulation of lipids to alleviate atherosclerosis in ApoE-/-mice through TLR4/NF-κB/NLRP3 signaling pathway.
