铁死亡干预结直肠癌细胞耐药的中西医研究进展

耐药是结直肠癌(colorectal cancer,CRC)晚期治疗的难点,而铁死亡(ferroptosis)作为一种不同于凋亡、坏死、自噬等传统途径的新兴死亡形式,能通过脂质过氧化的方式绕过CRC的耐药环节,进而诱导细胞死亡,为克服CRC耐药提供了新的干预靶点及治疗思路。详细论述铁死亡及CRC耐药的机制,探索相关干预靶点,从铁死亡干预化疗耐药、靶向耐药、免疫耐药等角度出发,总结近年来有关铁死亡干预CRC耐药细胞的研究进展,并概述了我国传统中药提取物诱导耐药CRC细胞铁死亡的近期研究,为后续更进一步的机制探索及临床研究打下基础。
Research Progress of Traditional Chinese and Western Medicine on Ferroptosis Overcome Drug Resistance of Colorectal Cancer
In the realm of colorectal cancer treatment,drug resistance is a formidable obstacle.Ferroptosis is a recently dis-covered mechanism of cell death that differs from necrosis,autophagy,apoptosis and other traditional pathways.It may trigger cell death through lipid peroxidation,which circumvents the drug resistance link in colorectal cancer(CRC)and provides a novel tar-get for intervention and therapeutic ideas to overcome drug resistance in CRC.This paper presented a comprehensive overview of the mechanisms underlying ferroptosis and CRC drug resistance,including relevant intervention targets and a summary of the latest advances in ferroptosis intervention research on drug-resistant CRC cells from the perspectives of ferroptosis intervention on im-mune,targeted and chemotherapeutic resistance.In addition,the relevant mechanisms of traditional Chinese medicine extracts in-tervening in drug-resistant colorectal cancer through ferroptosis were also summarized.This paper listed and summarized the tar-gets,pathways and processes of ferroptosis to overcome CRC drug resistance,laying the groundwork for more complex mechanisms and future clinical investigations.
