Study on Relationship between Flavonoid Formation and Element Content in Different Developmental Stages of Yinyanghuo[Epimedium sagittatum(Sieb.et Zucc.)Maxim.]Leaves
Objective To explore the changes of flavonoids and the accumulation of elements in the leaves of Yinyanghuo[Epi-medium sagittatum(Sieb.et Zucc.)Maxim.]at different developmental stages and to reveal the formation mechanism of fla-vonoids.Method The leaves of Yinyanghuo[Epimedium sagittatum(Sieb.et Zucc.)Maxim.]were divided into six stages from t1 to t6 according to different developmental stages,the contents of four flavonoids were determined by HPLC.The contents of to-tal flavonol glycosides were determined by UV spectrophotometry.The relative expression of 17 genes related to flavonoid biosyn-thesis key enzyme genes were determined by qRT-PCR method and the contents of 25 elements were determined by ICP-MS.Pearson correlation analysis was used to analyze the relationship between flavonoids and elements.Results With the development of leaves,the contents of epimedin A,B,C and total flavonol glycosides decreased significantly while the content of icariin in-creased at first and then decreased.However,the total amount of five flavonoids increased significantly and accumulated in t4 and t5 periods.The relative expressions were significantly up-regulated at t4 and t5,and decreased significantly at t2,the expression of different genes varied with leaf development.The contents of Mg,Ca,Ti,Mn,Fe,Sr and Ba increased significantly,while the contents of K and Co decreased significantly.There was a significant positive correlation between the contents of P,K,V,Cu and the contents of flavonoids in Yinyanghuo[Epimedium sagittatum(Sieb.et Zucc.)Maxim.].Conclusion The formation of fla-vonoids in Yinyanghuo[Epimedium sagittatum(Sieb.et Zucc.)Maxim.]leaves is affected by the degree of leaf development and the expression of related genes,and flavonoids is accumulated in the late stage of leaf development.
