Constructing Rat Model of Kidney-Yang Deficiency Type of Sarcopenia-Osteoporosis with Castration Method and Exploring The Mechanism
Objective Castration method was used to construct a rat model of kidney-Yang deficiency with sarcopenia-osteo-porosis and explore the correlation between its pathogenesis and musculoskeletal energy metabolism.Methods Sixty 3-month-old female SD rats were randomly divided into castration groups 1,2 and 3 and control groups 1,2 and 3,and then killed at 4,8 and 12 weeks after operation,respectively.The castration groups underwent bilateral oophorectomy,and the control groups under-went sham operation.During feeding,the animal behavior was observed and the body weight and grip strength were measured.The serum was collected to detect the levels of alkaline phosphatase(ALP),tartrate resistant acid phosphatase(TRAP),estradiol(E2)and irisin.The left hind limb triceps and left femur were collected for morphological detection and immunohistochemistry was used to observe irisin expression.The right quadriceps wet weight was weighed and the right femur bone density was detec-ted.Western Blot detection was performed to observe irisin expression.Results At 4 weeks after operation,the gripping power of rats in the castration groups was significantly decreased(P<0.05),and the expression of irisin in skeletal muscle and bone tissue of castration groups was significantly decreased by immunohistochemistry(P<0.05),but there was no significant difference in Western Blot among groups.At the 6th week after operation,kidney-Yang deficiency began to appear in the castration groups.At 8 weeks after operation,there was no significant difference in irisin expression in bone tissue between castration group and con-trol group(P>0.05).The serum TRAP content was significantly increased(P>0.05),and the remaining indexes were signifi-cantly decreased,and the morphological changes were in line with the trend.At 12 weeks after operation,only serum ALP content in the castration groups was significantly higher than that in the control groups(P<0.01),and the change trend of other indexes was the same as that 4 weeks ago.Conclusion Castration method can successfully construct the model of kidney-Yang deficiency type of sarcopenia-osteoporosis.The decreased expression of irisin in skeletal muscle of model animals is positively correlated with the development of the disease,and the decreased musculoskeletal energy metabolism may be one of the mechanisms of the disease.
