山楂提取物基于调控TBX21、GATA-3因子对血脂异常小鼠Th1、Th2细胞亚群的影响

目的 探究山楂提取物通过调控T-box转录因子21(TBX21)、GATA结合蛋白3(GATA-3)调节血脂异常小鼠Ⅰ型辅助性T细胞(Th1)、Ⅱ型辅助性T细胞(Th2)细胞亚群防治血脂异常机制研究。方法 60只C57BL/6J小鼠,随机分为6组,即正常组、高脂饮食组(High Fat Diet,HFD)、HFD+高剂量山楂提取物组、HFD+中剂量山楂提取物组、HFD+低剂量山楂提取物组、HFD+阿托伐他汀组,每组10只。HFD小鼠高脂饲料喂养8周,建立血脂异常模型。高、中、低山楂提取物组小鼠在高脂饲养4周后给予山楂提取物进行灌胃,阿托伐他汀组小鼠在高脂饲养4周给予阿托伐他汀进行灌胃。对照组和HFD组同时间口服灌胃等量生理盐水。4周后通过全自动生物化学分析仪检测血浆中甘油三酯(Triglyceride,TG),总胆固醇(Total Cholesterol,TC),高密度脂蛋白胆固醇(High Density Lipoprotein Cholesterol,HDL-C),低密度脂蛋白胆固醇(Low Density Lipoprotein Cholesterol,LDL-C)的含量;实时荧光定量 PCR(Quantitative Real-time-Polymerase Chain Reaction,qRT-PCR)检测各组小鼠脾脏组织 Tbx21、Gata3 mRNA 表达;蛋白免疫印迹法(Western Blot)检测各组小鼠脾脏组织Tbx21、Gata3蛋白表达;流式细胞术检测小鼠脾脏组织Th1、Th2细胞比例。结果 相较于正常组,高脂饮食能明显提高血清总胆固醇,甘油三酯和低密度脂蛋白胆固醇含量,小鼠脾脏Tbx21 mRNA及蛋白表达水平显著升高,Gata3 mRNA及蛋白表达水平显著降低,小鼠脾脏组织Th1细胞百分比显著上调,Th2细胞百分比显著下调;与HFD组相比,经山楂提取物及阿托伐他汀干预的血脂异常小鼠血清血脂得以改善,总胆固醇、甘油三酯和低密度脂蛋白胆固醇水平显著下降,高密度脂蛋白胆固醇水平显著上升,小鼠脾脏Tbx21 mRNA及蛋白表达水平显著降低,Ga-ta3 mRNA及蛋白表达水平显著提高,小鼠脾脏组织Th1细胞百分比显著下降,Th2细胞百分比显著上升,中药高剂量组与西药组调节相关因子的作用效果明显优于中药中、低剂量组,且对于HDL-C、LDL-C、Th1细胞比例、Tbx21、Gata3 mRNA及蛋白等指标的作用效果接近于西药组。结论 山楂提取物可以通过调控Tbx21、Gata3因子,进而影响Th1、Th2细胞分化过程,减轻脂质沉积,从而有效地防治血脂异常。
Effects of Shanzha(Crataegi Fructus)Extract on Th1 and Th2 Cell Subsets in Dyslipidemic Mice Based on Modulation of TBX21 and GATA-3 Factors
Objective To investigate the mechanism of Shanzha(Crataegi Fructus)extract in regulating T-box transcription factor 21(TBX21)and GATA-binding protein 3(GATA-3)to regulate the subpopulations of type Ⅰ helper T cells(Th1)and type Ⅱ helper T cells(Th2)in dyslipidemic mice to prevent and control dyslipidemia.Methods Sixty C57BL/6J mice were ran-domly divided into six groups,i.e.,normal control group,high fat diet(HFD)group,HFD+high dose Shanzha(Crataegi Fruc-tus)extract group,HFD+medium dose Shanzha(Crataegi Fructus)extract group,HFD+low dose Shanzha(Crataegi Fructus)extract group,and HFD+atorvastatin group,with 10 mice in each group.The HFD mice were fed with high-fat food for 8 weeks to establish dyslipidemia model.Mice in the high,medium and low dose Shanzha(Crataegi Fructus)extract groups were given Shanzha(Crataegi Fructus)extract by gavage after 4 weeks of high-fat feeding,and the mice in the atorvastatin group were given atorvastatin by gavage after 4 weeks of high-fat diet.In the control and HFD groups,the equal amounts of saline were adminis-tered orally by gavage at the same time,and the levels of plasma triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)were detected by an automated biochemistry analyzer af-ter 4 weeks of high-fat feeding.Quantitative Real-time-Polymerase Chain Reaction(qRT-PCR)was used to determine the expressions of Tbx21 and Gata3 mRNA.Tbx21 and Gata3 protein expressions were detected by Western Blot and the ratio of Th1 and Th2 cells in mouse spleen was detected by flow cytometry.Results Compared with the normal group,the high-fat diet signif-icantly increased the levels of serum TC,TG and LDL-C,the Tbx21 mRNA and protein expressions in the spleen of mice were significantly increased,the Gata3 mRNA and protein expressions were significantly decreased,the percentage of Th1 cells in the splenic tissues of mice was significantly up-regulated,and the percentage of Th2 cells was significantly down-regulated.Com-pared with the HFD group,the serum lipids of dyslipidemic mice intervened by Shanzha(Crataegi Fructus)extract and atorvasta-tin were improved,the levels of TC,TG and LDL-C were significantly decreased,HDL-C level was significantly increased,Tbx21 mRNA and protein expressions in the spleen of mice were significantly decreased,Gata3 mRNA and protein expressions were significantly increased,and the percentage of Th1 cells in mouse spleen tissues was significantly decreased and the percent-age of Th2 cells was significantly increased.The percentage of Th1 cells in the splenic tissues of mice decreased significantly,the percentage of Th2 cells increased significantly,and the effect of the Shanzha(Crataegi Fructus)extract high dose group and the HFD+atorvastatin group group in regulating the related factors was significantly better than that of the medium and low dose Shanzha(Crataegi Fructus)extract groups and the effect on the levels of HDL-C and LDL-C,the percentage of Th1 cells,the expressions of Tbx21,Gata3 mRNA and protein was close to that of the HFD+atorvastatin group group.Conclusions Shanzha(Crataegi Fructus)extract can effectively prevent and control dyslipidemia by regulating Tbx21 and Gata3 factors,which in turn affects the differentiation process of Th1 and Th2 cells and attenuates lipid deposition.
