Study on Activation Effect of Different Acupuncture Techniques on Skin Mast Cells in Eye Acupoint Area of MCAO/R Rats
Objective To explore the effect of different acupuncture techniques on the activation of mast cells in the eye acu-puncture point area of rats with cerebral ischemia-reperfusion(MCAO/R).Method Thirty-five rats were randomly divided into blank control group,model control group,no manipulation group,scraping handle group and twisting group.The MCAO/R model was created using an improved suture method,with intervention every 12 hours for 72 consecutive hours.The modified Garcia scoring method was used to score before and after intervention.Mast cells were stained with toluidine blue after euthanasia.The ELISA method was used to measure the levels of histamine(HA),substance P(SP),5-hydroxytryptamine(5-HT),prostaglan-din D2,tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in the acupoint area skin.Statistical analysis was con-ducted using SPSS 26.0.Results Compared with those of the model control group,the nerve function score of the no manipulation group,scraping handle group and twisting group decreased.The degranulation rate of mast cells increased,the levels of skin HA,SP,5-HT and PGD2 increased,the levels of serum TNF-α and IL-6 decreased,and the difference was statistically significant(P<0.01).The expressions of HA,SP,5-HT,PGD2,TNF-α and IL-6 in scraping handle group and twisting group were statistically significant(P<0.05).Conclusion The scraping or twisting method has a better effect on the activation of mast cells in the eye acupuncture area of MCAO/R rats than no manual manipulation.
