Study on Structural Changes of Polysaccharides in Huangqi(Astragali Radix)before and after Processing with Honey
The polysaccharides from raw and honey-processed Huangqi(Astragali Radix)were extracted by water extraction and alcohol precipitation.The polysaccharides were separated and purified by DEAE-52 chromatography and HW-65F gel col-umn.APS-1 and HAPS-1 were obtained from raw and honey-processed Huangqi(Astragali Radix),respectively.The purity and molecular weights of APS-1 and HAPS-1 were determined by high performance liquid gel permeation chromatography.The monosaccharides in polysaccharides were analyzed by electrochemical detector.The sugar chain structure of APS-1 and HAPS-1 was analyzed by infrared spectroscopy.The results show that APS-1 and HAPS-1 are the main polysaccharides in raw and honey-processed Huangqi(Astragali Radix).The molecular weights(Mw)of APS-Ⅰ and HAPS-Ⅰ were 7.839 kDa and 21.36 kDa,respectively.It is shown that APS-Ⅰ and HAPS-Ⅰ are consisted of arabinose,galactose,glucose,xylose,man-nose and levulose,in a molar ratio of 8.00∶3.00∶41.00∶1∶2.00 and 12.73∶3.68∶48.92∶1∶1.21,respectively.Both APS-1 and HAPS 1 have functional groups O-H,C-H and C=O.APS-1 also has functional groups C-O.The molecular weight of polysaccharide and the proportion of monosaccharide composition are changed after honey processing.This study can provide a reference for interpreting the synergistic effect of Huangqi(Astragali Radix)from the perspective of polysaccharide changes before and after processing.
