麦冬对肺炎支原体感染后咳嗽小鼠TRPA1、SP、CGRP的影响

目的 考察麦冬通过调节瞬时受体电位锚蛋白1(transient receptor potential A1,TRPA1)、P物质(substance P,SP)和降钙素基因相关肽(calcitonin gene related peptide,CGRP)表达对肺炎支原体感染后小鼠咳嗽的抑制作用。方法 通过分子相互作用(biomolecular interaction analysis,Biacore)技术检测麦冬与咳嗽因子TRPA1的亲合力,并采用体内药效学加以验证。实验分为空白组、模型组、喷托维林组、麦冬高剂量组、麦冬中剂量组、麦冬低剂量组,每组10只。除空白组外,其余5组BALB/c小鼠通过鼻腔滴入20 μL 106CCU肺炎支原体进行造模3 d,各组小鼠灌胃给予相应药物7 d。引咳实验观察各组小鼠3 min内咳嗽情况。苏木素-伊红(HE)染色法、Masson染色观察肺组织病理变化,免疫组化观察各组小鼠肺组织TRPA1、SP、CGRP蛋白表达,Real-time PCR检测小鼠肺组织TRPA1、SP、CGRP mRNA表达。结果 与模型组相比,麦冬高剂量组能显著降低小鼠咳嗽次数;HE和Masson染色结果显示,模型组支气管及肺泡均可见大量炎性细胞浸润,肺泡壁塌陷,而麦冬给药组肺组织病理改变均较模型组减轻;免疫组化和Real-time PCR结果显示麦冬高剂量组和中剂量小鼠肺组织TRPA1、SP、CGRP mRNA表达降低。结论 麦冬能够降低肺炎支原体感染后咳嗽小鼠肺组织TRPA1、SP、CGRP的表达从而达到治疗肺炎支原体感染后咳嗽。
Effect of Maidong(Radix Ophiopogonis)on TRPA1,SP and CGRP in Mice with Cough after Mycoplasma Pneumoniae Infection
Objective It investigated the inhibitory effects of Maidong(Radix Ophiopogonis)on cough in mice after Mycoplas-ma pneumoniae infection by regulating the expression of transient receptor potential A1(TRPA1),substance P(SP)and calcito-nin gene related peptide(CGRP).Methods The affinity of Maidong(Radix Ophiopogonis)with cough factor TRPA1 was detected by Biomolecular interaction analysis(Biacore)technique and verified by in vivo pharmacodynamics.The experiment was divided into blank group,model group,pentoxifylline group,Maidong(Radix Ophiopogonis)high dose group,Maidong(Radix Ophiopogo-nis)medium dose group and Maidong(Radix Ophiopogonis)low dose group,10 mice in each group.Except the blank group,the BALB/c mice of the remaining 5 groups were modeled by nasal drip of 20 μL 106 CCU mycoplasma pneumoniae for 3 d.The mice in each group were given the corresponding drugs by gavage for 7 d.Cough elicitation experiments were performed to ob-serve the coughing of mice in each group within 3 min.HE and Masson staining were performed to observe the pathological chan-ges of lung tissues and immunohistochemistry was performed to observe the expressions of TRPA1,SP and CGRP in the lung tis-sues of mice in each group.The mRNA expressions of TRPA1,SP and CGRP in the lung tissues of mice were detected by Real-time PCR.Results Compared with the model group,the Maidong(Radix Ophiopogonis)high dose group significantly reduced the number of coughs in mice.Hematoxylin-eosin(HE)staining and Masson staining showed that a large number of inflammatory cells infiltrated the bronchi and alveoli and collapsed the alveolar wall in the model group,while the histopathological changes in the Maidong(Radix Ophiopogonis)administration groups were reduced compared with those in the model group.Immunohisto-chemistry and Real-time PCR results showed that the expressions of TRPA1,SP and CGRP mRNA in the lung tissues of mice in the Maidong(Radix Ophiopogonis)high and medium dose groups decreased.Conclusion Maidong(Radix Ophiopogonis)was a-ble to reduce the expressions of TRPA1,SP and CGRP in the lung tissue of mice with cough after mycoplasma pneumoniae infec-tion,thus achieving the treatment of cough after mycoplasma pneumoniae infection.
