Study on Quantity Value Transfer of Standard Decoction in Raw and Processed Niuxi(Achyranthis Bidentatae Radix)
Objective To establish efficient high-performance liquid chromatography(HPLC)fingerprints for the standard de-coction of raw and processed Niuxi(Achyranthis Bidentatae Radix)and study the transfer of index components.Methods The chromatographic column was Venusil C18(250 mm×4.6 mm,5 μm)column.The column temperature was 30 ℃,and the detec-tion wavelength was 230,280 nm.The injection volume was 20 μL and the flow rate was 0.8 mL·min-1.Chromatographic Fin-gerprint Similarity Evaluation System of Traditional Chinese Medicines(2012 Edition)was used to evaluate the similarity and de-termine the common peak.The contents of index components were determined,and the transfer rate and extraction rate were cal-culated.Results The similarity of all 45 batches of samples was above 0.934.Forty-five batches of samples were calibrated with 10 common peaks,referring to the recognition of 7 peaks,namely 5-hydroxyfurfural,polypodine B,β-ecdysone,25 R-inokos-teron,25S-inokosteron,ginsenoside R0 and chikusetsu saponin Ⅳa.The average transfer rates of the seven components in the standard decoction of the raw and processed products of Niuxi(Achyranthis Bidentatae Radix)were 291.53%,28.90%,56.74%,54.61%,53.01%,64.28%,61.87%and 192.21%,61.16%,56.10%,54.69%,51.80%,58.06%,43.70%and 111.29%,53.59%,62.47%,60.84%,57.77%,50.20%,48.69%.The average extraction rate of the standard decoction of the raw and processed products of Niuxi(Achyranthis Bidentatae Radix)of standard decoction was 35.06%,34.02%and 36.32%.Conclusion The fingerprint method is stable and reliable,and it can fully demonstrate the transfer of index components during sample preparation,which can provides a reference for the study of classic prescriptions and compound preparation contai-ning Niuxi(Achyranthis Bidentatae Radix).
