Icariside Ⅱ inhibits intimal hyperplasia by down-regulating the TGF-β1/Smads signaling pathways in the rat carotid artery after balloon injury
Objective To research the effects of Icariside Ⅱ (ICSⅡ) on vascular intimal hyperplasia in balloon-injured carotid artery of rats and further explore wheter the regulating mechanisms are associated with TGF-β1/Smads signaling pathways.Methods Male rats were randomly divided into the sham operation group and the carotid artery balloon-injured groups.Animal carotid artery models were established by balloon injury,and the survival rats were randomly divided into three groups:model,ICSⅡ-L,ICSⅡ-H groups.ICSⅡ-L and ICSⅡ-H groups were administered with ICSⅡ (10,20mg/kg · d),qd) by gavage for 14 days.The same volume of distilled water was administered in sham group and model group for 14 days.Twenty-four hours after the last dose,rats were anesthesized,the injured left common carotid artery was collected,the neointimal hyperplasia levels were observed in the injured side of the carotid vessel with H.E.staining.The protein expressions of TGF-β1,p-Smad2,p-Smad3 and MMP-2 in rat carotid were examined by Westem Blot.Results Compared with the sham group,the neointimal hyperplasia was evident in model group after balloon injury.In addition,it was found that the protein expressions of TGF-β1,p-Smad2,p-Smad3 and MMP-2 were increased in carotid artery of model group (P <0.05).However,ICSⅡ could attenuate the thickness of neointima and decrease the elevated protein expressions of TGF-β1,p-Smad2,p-Smad3 and MMP-2 after 14 days of treatment on balloon injuriedrat carotid (P <0.05).Conclusion ICSⅡ can significantly attenuate the intimal hyperplasia in rat carotid arteries induced by balloon-injury,and the regulating mechanisms may be associated with down-regulating the TGF-β1/Smads pathways.
