Analysis on the Medication Rules of Traditional Chinese Medicine in the Treatment of Postoperative Gastrointestinal Dysfunction of Gastric Cancer based on Data Mining and Network Pharmacology
Objective To analyze the medication rules of traditional Chinese medicine(TCM)in the treatment of postoperative gastrointestinal dysfunction(POGD)of gastric cancer based on data mining and network pharmacology.Methods Relevant literature was collected and screened in CNKI,Wanfang,VIP,CBM and to be processed with standardization.Excel 2019 was used to perform frequency statistics on medication,nature,flavor,and meridian tropism.IBM Modeler 18.0 software was used for association rule analysis of drugs,and IBM SPSS Statistics 26.0 software was used for cluster analysis of high-frequency drugs.Drug-active ingredient-target network and drug-disease key target protein-protein interaction(PPI)network were constructed.Gene ontology(GO)enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis were performed.Results A total of 61 prescriptions were included,involving 118 herbs.The top four herbs with highest frequency were Largehead Atractylodes Rhizome,Glycyrrhizae Radix et Rhizoma,Citri Reticulatae Pericarpium,Poria.The distribution of four nature was mainly based on warm,combined with mild and cold.The distribution of five flavors was mainly based on sweet,combined with bitterness and pungent.The frequency distribution of meridian tropism was mainly based on spleen,followed by lung and stomach.The analysis of association rules showed that there were 13 pairs of couplet medicines with strong association,such as"Largehead Atractylodes Rhizome-Poria"and"Largehead Atractylodes Rhizome-Citri Reticulatae Pericarpium".High frequency drugs could be classified into three categories.A total of 26 potential active ingredients with 362 targets as well as 137 key targets of drug-disease intersection were selected from Largehead Atractylodes Rhizome-Poria.Tumor necrosis factor(TNF),peroxisome proliferator activated receptors γ(PPARG),and prostaglandin endoperoxide synthase 2(PTGS2)were selected as key targets,while phosphoinositide 3-kinase-protein kinase B(PI3K-AKT)and mitogen activated protein kinase(MAPK)were considered as the potential signaling pathways of core drug combination in the treatment of POGD.Conclusion TCM treatment for POGD of gastric cancer mainly focuses on warming and tonifying from the perspective of spleen and lung,emphasizing the regulation of qi activity,and underlining both symptoms and root causes.Largehead Atractylodes Rhizome-Poria may be the core drug combination for treating POGD of gastric cancer.
