乙酸乙酯重结晶纯化植物甾醇的工艺研究

植物甾醇是一种天然活性物质,具有广阔的发展前景和开发价值.为提高植物甾醇产品纯度,优化乙酸乙酯重结晶纯化植物甾醇的工艺.以粗甾醇为原料,乙酸乙酯为结晶溶剂,研究结晶终温、降温速率、搅拌速率及结晶次数对总甾醇含量及收率的影响,并对植物甾醇产品相应的质量指标进行分析.结果表明:在结晶初始温度 50℃、降温速率 0.6℃/min、结晶终温 23℃、搅拌速率 46 r/min、结晶 3 次的条件下,总甾醇含量可达 99.10%,总甾醇收率为 80.34%;产品质量指标检测结果分别为酸价0.49 mg/g,熔点 139℃,灰分 0.15%,水分含量 0.16%,符合相关标准.研究结果可为生产高纯度植物甾醇提供技术支持.
Purification of phytosterol by recrystallization with ethyl acetate
Phytosterol is a kind of natural active substance,which has broad development prospects and val-ue.With the upgrading of people's health consumption and the rapid expansion of the functional nutrition market,the market demand of phytosterol industry has also increased.Therefore,how to improve the puri-ty of phytosterol to meet the application market needs was an urgent problem to be solved.In this article,crude sterol was used as raw material and ethyl acetate as crystallization solvent.The optimum crystalliza-tion conditions were obtained by single factor experiments and response surface experiments.The results showed that the optimal crystallization conditions were obtained as follows:crystallization temperature 23℃,cooling rate 0.6℃/min,stirring rate 46 r/min,and crystallization times 3.Under these condi-tions,the total sterol content could reach 99.10%,and the yield was 80.34%.The theoretical analysis re-sults of the response surface were consistent with the predicted values,indicating that the model was relia-ble.Combined with the actual production,the recovered mother liquor was recycled in the same way as a-bove.The corresponding quality indicators of refined sterol based on relevant standards were analyzed.The sterol crystal form was loose powder without,irritating odor after repeated crystallization.The physical and chemical properties of refined sterol were obtained as follows:acid value 0.49 mg/g,melting point 139℃,ash content 0.15%,which met the corresponding standards.The purity of phytosterol can be sig-nificantly improved by recrystallization of ethyl acetate,and the corresponding quality characteristics have been better improved.This study is expected to provide a research basis for subsequent high value utiliza-tion of phytosterol.
