AflbasR基因对储粮霉菌黄曲霉生长发育和毒素合成的影响

MYB 类型的转录因子参与调控生物生长发育、代谢和对外界胁迫反应等,为研究 MYB 型转录因子在黄曲霉中的功能,通过同源重组的方法构建转录因子 AflbasR 缺失菌株,分析其对黄曲霉生长、分生孢子产量、黄曲霉毒素 B1(AFB1)合成、耐受性和致病性等的影响.结果表明:与对照菌株相比,ΔAflbasR 菌株生长速度缓慢,分生孢子头密集且大,分生孢子梗短而粗;AflbasR 基因的缺失导致黄曲霉对细胞壁胁迫试剂刚果红以及氧化胁迫试剂(H2O2)的耐受力显著减弱;AFB1 的生物合成量显著下降,对花生种子的感染能力降低.本研究丰富了黄曲霉毒素生物合成调控理论,为预防黄曲霉污染提供潜在靶点.
Effects of AflbasR gene on the growth,development and aflatoxin biosynthesis of the grain storage fungus Aspergillus flavus
MYB-type transcription factors are involved in regulating biological growth and development,metabolism,and responses to external stresses.To investigate the function of MYB-type transcription fac-tors in Aspergillus flavus(A.flavus),a deletion mutant of the transcription factor AflbasR-deficient strain was constructed through homologous recombination,and its effects on the growth,conidial production,aflatoxin B1(AFB1)synthesis,stress tolerance,and pathogenicity of A.flavus were analyzed.The results showed that compared with the control strain,the ΔAflbasR strain exhibited slower growth,denser and lar-ger conidial heads,and shorter and thicker conidial peduncle.The deletion of AflbasR gene significantly re-duced the tolerance of A.flavus to the cell wall stress agent Congo Red and oxidative stress(H2O2).The biosynthesis of AFB1 was significantly decreased,and the ability to infect peanut seeds was reduced.This study enriches the theory of aflatoxin biosynthesis regulation and provides potential targets for preventing A.flavus contamination.
