珍稀濒危观赏竹种筇竹遗传多样性研究

对筇竹Qiongzhuea tumidinoda居群间及居群内遗传多样性进行了研究.通过基因组多态性分析,筛选出13个随机引物对筇竹样品进行扩增,得到清晰可重复且多态性明显的条带,多态位点率为76.38%.在NTSYS程序中,统计这些扩增产物用来计算遗传相似性系数和遗传距离,建立UMPGA(不加权算术平均法)聚类图,得出筇竹居群间的基因多样性大于居群内的结论,在此基础上,探讨筇竹保护对策和建议.
Genetic Diversity of the Rare and Endangered Species Qiongzhuea tumidinoda
The genetic diversity within and among populations of Qiongzhuea tumidinoda was analyzed using RAPD markers.Through polymorphic genome analysis,13 random primers were selected to amplify samples of Q.tumidinoda,clear reproducible and polymorphic bands were resulted in,and the rate of polymorphic loci was 76.38%.Using NTSYS-PC program,Nei's genetic similarity coefficients were generated and the UMPGA dendrogram was constructed.It was concluded that the genetic diversity between populations was higher than that of within populations.At last,some suggestions on the conservation of Q.tumidinoda were made.
