    Effect of Sishen pill on expression of type Ⅰ and type Ⅲ interferon in acute ulcerative colitis mice model

    查看更多>>摘要:Objective:To investigate the effects of Sishen pill on the expression of type I interferon(IFN) and type Ⅲ interferon and their receptors in colonic tissues of mice with acute ulcerative colitis(UC). Methods: Male C57BL/6Cnc mice were randomly divided into control group, model group, sishenwan group and salazosulfapyridine group. The model was made with 0.2 mL 4% dextran sodium sulfate (DSS) for 5 days, and the control group was given 0.2mL normal saline by gavage. On the second day of modeling, sishen pill group was given 0.2mL 1.5 g·kg-1 sishen pill, and SASP group was given 0.2mL 0.25 g·kg-1 sulfasalazine, twice a day, for 7 days. During the administration period, the disease activity index (DAI) of mice was calculated every day. After administration, the histopathological changes of colon tissues of mice in each group were observed by hematoxylin eosin (HE) staining, and the histological scores were calculated. The expression of IFN-α, IFN-β, IFN-λ2 and IFN-λ3 mRNAs in colon tissues of mice in each group were detected by qRT-PCR. The expression levels of IFN-α, IFN-β, IFN-λ2 and IFN-λ3 in colon tissues of mice in each group were detected by ELISA. Western blot was used to detect the expression of interferon receptors IFNAR1, IFNAR2 and IFNLR1 in colon tissues of mice in each group. Results: Compared with the control group, the DAI of mice increased significantly (P<0.001) in the model group. The inflammatory cells in colonic tissues infiltrated heavily, lymph nodes enlarged, colonic mucosal structure destroyed, crypt structure lost, inflammation involved a wide range, and the histological score increased significantly (P<0.001). The levels of IFN-α, IFN-β and IFN-λ2 mRNA were significantly decreased (P<0.05, P<0.05, P<0.01). The expression levels of IFN-α, IFN-β, IFN-λ2 and IFN-λ3 were significantly decreased (P<0.01, P<0.01, P<0.001, P<0.001). The levels of IFNAR1, IFNAR2 and IFNLR1 were significantly decreased (P<0.01, P<0.05, P<0.01). Compared with the model group, the DAI decreased significantly (P<0.001) in Sishen pill group, the infiltration of inflammatory cells in colon tissue were significantly reduced, the structural regeneration of colon mucosa was significantly recovered, the crypt structure was significantly recovered, the lymph nodes were significantly reduced, the range of inflammation involvement was reduced, and the histological score was significantly reduced (P<0.001). The levels of IFN-α, IFN-β and IFN-λ2 mRNA were significantly increased (P<0.01, P<0.001, P<0.001). The levels of IFN-α, IFN-β, IFN-λ2 and IFN-λ3 were significantly increased (P<0.01, P<0.001, P<0.01, P<0.001). The levels of IFNAR1, IFNAR2 and IFNLR1 were significantly increased (P<0.05, P<0.001, P<0.05). Conclusion: Sishen pill may alleviate the symptoms and signs of mice with acute ulcerative colitis by regulating the expression of type I and type Ⅲ interferon and their receptors in colon tissues.

    Effects of compatibility of Scutellaria baicalensis stems and Polygonum cuspidatum on TRPV1 expression and inflammatory cytokines in rats with acute lung injury

    查看更多>>摘要:Objective:The protective effect of Scutellaria baicalensis Stems and Polygonum Cuspidatum compatibility on lipopolysaccharide (LPS)-induced acute lung injury (ALI) in rats was studied by observing the expression of TRPV1 and inflammatory cytokines. Methods: 48 male SD rats were randomly divided into 6 groups: control group, model group, dexamethasone group (5mg / kg) and Scutellaria baicalensis Stems -Polygonum Cuspidatum (3.5, 7 and 14g / kg). The administration group was gavaged for 7 days, and the control group and model group were given the same amount of 0.9% sodium chloride. On the 8th day, except the control group, rats in other groups were injected with 8mg / kg LPS through caudal vein to induce Ali model. Take the rat lung tissue 6 hours after modeling, and calculate the wet/dry weight ratio (W/D) of the rat lung tissue; HE staining to observe the pathological changes of lung tissue; Determine the content of tumor necrosis factor-α (TNF-α) and interleukin-1β (1L-1β) in alveolar lavage fluid (BALF) and the activity of superoxide dismutase (SOD) in serum; Detect the mRNA and protein expression levels of TRPV1 receptor in rat lung tissue. Results: Compared with the model group, Scutellaria baicalensis Stems -Polygonum Cuspidatum can significantly reduce the damage of lung tissue structure and bleeding state, W/D value and TNF- α、 IL-1 β The content of TRPV1 decreased, the level of SOD increased, and the mRNA and protein expression of TRPV1 receptor decreased. Conclusion: The combination of Scutellaria baicalensis Stems -Polygonum has a protective effect on acute lung injury in rats, and its mechanism may be related to down-regulating the expression of TRPV1 and inhibiting the levels of TNF-α and IL-1β in inflammatory cells.

    An exploration on the protective mechanism of Xuduan Zhongzi prescription against epididymis oxidative damage in oligoasthenospermia model rats based on Nrf2-NQO1/γ-GCS signaling pathway

    查看更多>>摘要:Objective: To investigate the protective mechanism of Xuduan Zhongzi prescription against epididymal oxidative damage in oligoasthenospermia model rats. Methods: Forty SD rats were randomly divided into blank group, model group, Xuduan Zhongzi prescription group (10g/kg)and L-carnitine group(0.1g/kg). Except blank group, all induced oligoasmospermia. The blank group and model group were given normal saline intragastric administration, the Xuduan Zhongzi prescription group was given Xuduan Zhongzi prescription solution intragastric administration, and the L-carnitine group was given L-carnitine intragastric administration. HE staining was used to observe the epididymis structure after 8 weeks. The concentration and activity rate of epididymis sperm were measured by sperm quality. MRNA and protein expression levels of Nrf2, NQO1 and γ -GCs in epididymis were detected by RT-qPCR and immunohistochemistry. Results: ①HE staining: in the blank group, the epididymis tubes were arranged tightly and regularly, the tissue structure was complete, the epithelial cells were arranged orderly, and the lumen sperm were numerous and evenly distributed. The epididymis of model group showed structural atrophy, loose arrangement, enlarged mesenchyme, increased cell debris and significantly reduced sperm cells. Compared with the model group, the lumen lesions of epididymis in Xuduan Zhongzi prescription group and L-carnitine group were significantly improved, and the amount of normal sperm in lumen was increased and the distribution was uniform. ② Results of sperm quality comparison among each group: sperm density and sperm motility rate: compared with blank group, sperm density and sperm motility rate in other groups were significantly decreased (P < 0.05), and sperm density and sperm motility rate in model group were significantly decreased (P < 0.05); Compared with model group, the sperm density and motility rate in Xuduan Zhongzi prescription group and L-carnitine group were significantly increased (P < 0.05). ③RT-qPCR and immunohistochemistry: Compared with the blank group, the mRNA and protein levels of Nrf2, NQO1 and γ -GCs in epididymal rats in model group were significantly decreased (P<0.05), while the mRNA and protein levels of Nrf2, NQO1 and γ -GCs were significantly increased in L-carnitine group and Continua seed formula group (P<0.05). Conclusion: Xuduan Zhongzi prescription can reduce oxidative stress damage and improve sperm quality of oligoasthenospermia. The mechanism may related to promoting the activation of Nrf2-NQO1/γ-GCS pathway in epididymis of oligoasthenospermia rats, and up-regulate the expressions of Nrf2, NQO1 and γ-GCS proteins.

    Correlation study between dendritic cell and eosinophil in refractory rhinosinusitis

    查看更多>>摘要:Objective: To investigate the correlation of refractory rhinosinusitis by detecting the expression of dendritic cells and eosinophils infiltration in nasal mucosa. Methods: Histological staining was performed to detect eosinophils infiltration respectively in 30 normal nasal mucosa, 30 chronic rhinosinusitis tissues and 30 refractory rhinosinusitis tissues,Immunohistochemistry and immunofluorescence double labeling were performed to detect the expression of CD83 and CD11c in each group, and univariate anova was used to analyze the differences among groups.The correlation between the co-expression of CD83 and CD11c and the infiltration of eosinophilic cells was measured by Pearson correlation analysis. Results: The positive expression of dendritic cells immune markers CD83 and CD11c and eosinophils infiltration of refractory rhinosinusitis was significantly increased compared with the other two groups (P <0.05). Through Pearson correlation analysisthe, correlation coefficient r between CD83 and CD11c co-expression and eosinophils infiltration in each group was positive and P <0.05, both were positively correlated. Conclusion: The number of dendritic cells in refractory rhinosinusitis increased significantly and was positively correlated with the infiltration of eosinophils, which was involved in the development of refractory rhinosinusitis.

    Effects of post-dilation on coronary blood flow and MACE events following primary percutaneous coronary intervention in patients with STEMI

    查看更多>>摘要:Objective: This retrospective cohort study aimed to evaluate the effect of post-dilation on coronary blood flow and MACE events during hospitalization and 1 year follow-up following primary PCI in patients with ST-segment. Methods: 419 eligible patients who underwent PPCI due to STEMI between January 2015 and October 2019 were enrolled. The CTFC, final QCA, and the incidence of no-reflow/slow-flow during different procedure moments were assayed. Study end points was to compare two groups of patients with clinical characteristics, compared two groups of patients with the incidence of no-reflow and slow-flow, and the incidence of MACE during hospitalization and 1-year follow-up. Results: The incidence of final no-reflow/slow-flow in the post-dilation group was not significantly higher than that in the non-post-dilation group (24.3% vs.19.4%; p = 0.238).There was no significant statistical difference in MACE events during hospitalization, but for the 1-year follow-up, the incidence of Target vessel revascularization and Target lesion revascularization in the post-dilation group was lower than that in the non-post-dilation group. A multivariable logistic regression model revealed that age (OR=1.078, 95%CI=1.038-1.120; P <0.001), history of diabetes (OR=3.009, 95%CI=1.183-7.654; P =0.021), post-dilation (OR=0.192, 95%CI=0.067-0.549; P=0.002) were independently correlated with long-term follow-up of MACE.Conclusion: Post-dilation does not increase poor prognosis during hospitalization, and reduces the incidence of TVR and TLR events during long-term follow-up.

    Effect of total glucosides of paeony on serum cytokines in patients with psoriasis and its clinical efficacy: A meta-analysis

    查看更多>>摘要:Objective: To evaluate the relationship between the changes of inflammatory cytokines and clinical efficacy in patients with psoriasis treated with Total glucosides of paeony by Meta analyses, and to explore the microcosmic mechanism of effective treatment of psoriasis with Total glucosides of paeony from the point of view of evidence-based medicine. Methods: The databases of CNKI, Wanfang, VIP, SinoMed, PuMed, Embase and Cochrane Library were searched by computer, and the time range was from the establishment of the database to May 2020. After screening, data extraction and bias risk assessment, Revman5.3 software was used for statistical analysis. Results: A total of 998 patients were included in 11 clinical studies, including 502 patients in the trial group and 496 patients in the control group. The results of Meta analysis showed that there was significant difference in the expression of cytokines between the two groups (MD=-10.97, 95%Cl[-15.56,-6.37]). The effective rate of treatment (OR=3.57, 95%Cl[2.58, 4.94]) and the decrease of PASI score (MD=-4.55, 95%Cl[-5.91,-3.20]) in the combined use of Total glucosides of paeony group were superior to those in the control group. Conclusion: Total glucosides of paeony can regulate immune and inflammatory response and improve skin lesions by inhibiting the expression of cytokines such as IL-2, IL-8, IL-23 and TNF-α in serum of patients with psoriasis. In view of the low overall quality of the included studies, larger samples and higher quality clinical trials are still needed to obtain more sufficient evidence.

    Efficacy of the method of tonifying Qi, resolving phlegm and eliminating stasis in thetreatment of AECOPD: A meta analysis

    查看更多>>摘要:Objective: To systematically evaluate the efficacy and safety of the method of tonifying qi, resolving phlegm and eliminating stasis in the treatment of acute exacerbation of chronic obstructive pulmonary disease. Methods: Search the China Knowledge Network (CNKI), VIP Chinese Science and Technology Journal Database (VIP), Wanfang Data Knowledge Service Platform, China Biomedical Literature Database (CBM), PubMed, Embase, Cochrane Library, and select the databases that meet the requirements for tonifying qi, resolving phlegm and eliminating stasis. The stasis method in the treatment of AECOPD was included in the standard literature, and RevMan 5.4 software was used for statistical analysis. Results:A total of 10 articles were included, with a total of 854 patients, 435 in the treatment group and 419 in the control group. Systematic analysis shows that the method of tonifying qi, resolving phlegm and eliminating stasis combined with conventional Western medicine treatment of AECOPD is significantly different in total clinical effective rate than conventional Western medicine treatment alone [OR=3.98,95%CI(2.23,7.11), P<0.00001];In terms of lung function, FEV1/FVC[MD=6.08,95%CI(5.01,7.15), P<0.00001], FEV1[ MD=0.07,95%CI(0.01, 0.13),P=0.02], FEV1%[MD=4.56,95%CI(3.09,6.02),P<0.00001] is significantly higher than the control group; arterial oxygen partial pressure (PaO2)[MD=18.47,95%CI(16.77,20.16), P<0.00001]Significant improvement compared to the control group; arterial partial pressure of carbon dioxide (PaCO2) [MD=-7.48,95%CI(-8.7,-6.26), P<0.00001] was significantly lower than the control group. Conclusion: The current evidence shows that the method of tonifying qi, resolving phlegm and eliminating stasis combined with conventional Western medicine treatment of chronic obstructive pulmonary disease in acute exacerbations can improve clinical efficacy and improve the quality of life of patients.

    查看更多>>摘要:Objective: To analyze the potential targets and mechanism of Xingxiao Pill for the treatment of lung cancer based on network pharmacology and molecular docking, providing reference for the research and clinical development of Xingxiao Pill. Method: The main chemical constituents and their targets of Xingxiao Pill was obtained through BATMAN-TCM database. Effective compounds were screened based on SwissADME and their targets were predicted by SwissTargetPrediction. Main lung cancer targets were obtained and integrated through GeneCards, OMIM(OMIM-Gene-Map-Retrieval), TTD and DRUGBANK databases. The intersection targets of Xingxiao Pill for lung cancer were obtained by Venn. The String platform was used to obtain the intersection targets. The core targets of Xingxiao Pill for lung cancer were screened by BisoGenet3.0. The Metasacpe database was used to analyze the biological processes and pathways involved in intersection targets. Cytoscape3.7.2 software was used to construct the "active ingredients of Xingxiao Pill-targets-pathways" network. Finally, Autodock was used for molecular docking verification. Results: The core active components of Xingxiao Pill for the treatment of lung cancer were morin, testosterone, 17-Beta-Estradiol, alpha-Estradiol, muscone, commiferin, commisterone, octyl acetate, etc. The primary pathways mainly included PI3K/Akt, MTOR, MAPK signaling pathway and MicroRNAs in cancer, etc. It has been proved that the combination of testosterone and estradiol with AKT1, SIRT7 and MDM2 was stable and could be used as reference. Conclusion: This study preliminarily revealed the mechanism of Xingxiao Pill on treating lung cancer with multiple components, targets and pathways, providing some ideas and references for clinical application and basic research in the future.

    Study on the mechanism of "Salvia chinensis and Radix Ranunculi Ternati" drug pair in the treatment of lung cancer

    查看更多>>摘要:Objective: To explore the molecular biological mechanism of the "salvia chinensis and radix ranunculi ternati" drug pair in the treatment of lung cancer based on network pharmacology. Methods: Searching the TCMSP database and previous literatures to screen the active compounds which resist lung cancer activity in salvia chinensis and radix ranunculi ternati. The candidate compounds were unified in the DrugBank to find the corresponding drug targets which were corrected to the standard gene names by the UniProt database. The SwissTargetPrediction platform was used to predict other targets. Searching GeneCards, OMIM and DrugBank to obtain genes related to lung cancer. After taking the intersection, the candidate gene target of drug pair in the treatment of lung cancer could be obtained. The "herbs-compounds-targets-disease" network was bulit with Cytoscape, and the PPI network was bulit on the STRING platform while the core network nodes were screened. GO and KEGG analysis on candidate genes was implemented through Metacape platform, and a "pathways-targets" network was bulit to further screen key genes. Results: A total of 16 active compounds in salvia chinensis, 18 active compounds in radix ranunculi ternati, 164 candidate targets, 2443 GO functions and 170 KEGG pathways was obtained. Conclusion: The effective compounds of " salvia chinensis and radix ranunculi ternati " drug pair in the treatment of lung cancer are quercetin, ursolic acid, β-sitosterol and caffeic acid. The key targets are MAPK1, AKT1, PIK3R1, RAF1 and EGFR. GO functions mainly include cytokines, oxidative stress, plasma membrane transmission, protein kinase binding and activity, apoptosis. KEGG could directly regulate pathways in cancer, non-small cells lung cancer pathway and small cell lung cancer pathway. KEGG also involves EGFR tyrosine kinase inhibitor resistance, IL-17, TNF, PI3K-AKT signaling pathway and apoptosis. This study reveals the molecular biological mechanism of "salvia chinensis and radix ranunculi ternati" drug pair in the treatment of lung cancer. It is reasoned that its potential targets affect multiple signaling pathways and ultimately resist the proliferation, differentiation, invasion, metastasis and promote apoptosis of lung cancer cells. Evidence for further experimental study is provided by this study.

    Research progress of Taohong Siwu Decoction and its modified prescription in the treatment of diabetic peripheral neuropathy

    查看更多>>摘要:Western medicine mainly focuses on the conventional basic treatment of Diabetic peripheral neuropathy (DPN), such as blood glucose control, anti-oxidative stress, and nerve nutrition, but the efficacy and safety of the drugs cannot be guaranteed.As an important prescription for promoting blood circulation, removing blood stasis and nourishing blood, Taohong Siwu Decoction can not only promote cell proliferation and repair, reduce inflammatory factors in the blood, reduce the whole blood viscosity and plasma viscosity, and has a good anticoagulation effect.Moreover, Taohong Siwu Decoction and its added or subtracted prescription, foot bath and combined acupuncture have the effect of regulating blood sugar, improving nerve conduction velocity and relieving clinical symptoms, without obvious adverse reactions.This article reviews Taohong Siwu Decoction and its addition and subtraction in the treatment of diabetic peripheral neuropathy.
