    Study on the mechanism of Haizao Yuhu Decoction in inhibiting pro-liferation of thyroid papillary carcinoma cells

    查看更多>>摘要:Objective: To clarify the inhibitory mechanism of Haizao Yuhu Decoction on BRAFV600E mutation-driven papillary thyroid carcinoma cells. Methods: Prepare seaweed Yuhu Decoction medicated serum, select human normal thyroid cells Nthy-ori3-1 as the normal control group, and PLX4032 as the positive control drug. The experiment was divided into normal control group, model control group, PLX4032 group, Haizao Yuhu Decoction group (referred to as HYD group), Haizao Yuhu Decoction group + PLX4032 (referred to as HYD+PLX4032 group), and high iodine water group, at 8h and 24h respectively. At 72h, the cell proliferation of each group was detected by MTT method; at 24h, the expression of ERK and p-ERK protein in each group was detected by Western blot. Results: The longer the time, the more obvious the inhibitory effect of Haizao Yuhu Decoction-containing serum on the proliferation activity of BCPAP cells: Compared with the normal control group, the BCPAP cell proliferation activity of the model control group was significantly enhanced at 24h and 72h (P <0.05); At 24h, the PLX4032 group, HYD group, and HYD+PLX4032 group all showed a tendency to inhibit the proliferation of BCPAP cells, but there was no statistical difference; at 72h, compared with the model control group, the PLX4032 group The cell proliferation activity of the HYD group and HYD+PLX4032 group was significantly inhibited (P <0.05), and there was no difference between the high iodine water group and the model control group (P>0.05). In inhibiting the proliferation of BCPAP cells, the medicated serum of Shanghai Zaoyuhu Decoction has a synergistic effect with PLX4032 (F=10.87, P=0.005). Western blot results showed that there was no difference in the expression of ERK1/2 protein between the groups, but there were significant differences in the expression of p-ERK1/2 protein between the groups: Compared with the normal control group, the expression of p-ERK1/2 protein in the model group increased significantly (P < 0.05); Compared with the model control group, the expression of p-ERK1/2 protein in the PLX4032 group, the HYD group, and the HYD+PLX4032 group decreased significantly (P <0.05), and there was no difference between the high iodine water group and the model control group (P> 0.05). Conclusion: The medicated serum of Haizao Yuhu Decoction has an inhibitory effect on the proliferation of BCPAP cells, and its mechanism may be inhibiting the proliferation of BCPAP cells by inhibiting ERK1/2 protein phosphorylation, a protein post-translational modification process; the medicated serum of Haizao Yuhu Decoction may have Help enhance the effect of PLX4032 curative effect.

    Therapeutic effect and mechanism of Shenqi Zhilong Decoction on mice with membranous nephropathy through ERK/cPLA2 signaling pathway

    查看更多>>摘要:Objective: To explore the therapeutic effect and underlying mechanism of Shenqi Zhilong Decoction on mice with membranous nephropathy (MN). Methods:Mice with MN was established by injecting cationic bovine serum albumin (c-BSA) into tail vein for several times. model mice were randomly divided into MN group (equal amount of distilled water), Shenqi Zhilong Decoction low dose group (12 g crude drug/kg), Shenqi Zhilong Decoction high dose group (24 g crude drug/kg), and Tripterygium wilfordii polyglycoside tablet group (14 mg/kg). Another 10 un-treatment mice were taken as control group (equal amount of distilled water). The drug was administered orally once a day for 4 weeks. After the last administration, 24 hours urine was collected to determine the urinary protein content; blood from inner canthus was collected to measure the changes of kidney function, liver function, blood lipid and levels of IL-6, IL-4 and TNF-α in serum in each group; HE staining was used to observe the pathological changes of kidney. Immunohistochemical staining was used to observe the expression of IgG in kidney. The protein expression of ERK1/2 and cPLA2 in renal tissues was determined by Western-blot method. The gene expression of Neph1, Nephrin and Podocin mRNA in kidney tissues were detected by RT-PCR. Results: Compared with model group, Shenqi Zhilong decoction at low-dose and high-dose could significantly reduce the value of urine protein in MN mice; Decreased TC and TG levels (P<0.05 or P<0.01); Increased the levels of ALB and TP in liver function (P<0.05 or P<0.01); has no significant effects on the levels of CRE, UREA and UA in renal function (P>0.05). Decreased the contents of IL-6, IL-4 and TNF-α in serum (P<0.05 or P<0.01); Significantly down-regulated the protein expression levels of p-ERK1/2 and p-cPLA2 in kidney tissues of MN mice (P<0.05 or P<0.01); Significantly increased the expression levels of NephP1, Nephrin and Podocin mRNA in renal tissues (P<0.01). Conclusion: Shenqi Zhulong Decoction has a good therapeutic effect on MN mice, and the mechanism of action is related to regulate the expression of related genes of Nephrin-Podocin-Neph1 receptor complex for protecting the glomerular filtration barrier, and inhibite the activation of ERK/cPLA2 pathway for relieving damage of GEC and reduceing secretion of pro-inflammatory cytokines.

    查看更多>>摘要:Objective: The relA gene,a gene associated with the synthesis of rigorous response signaling molecule (p)ppGpp in Burkholderia pseudomallei, was knocked out and its effects on the biological functions of bacterial growth, motility and biofilm formation were investigated. Methods: The plasmids with trimethoprim resistance (TPR) were modified by enzyme digestion and enzyme ligation method. The TPR fragment was linked to the suicide plasmid pK18mobSacB containing SacB sucrose killing gene by Bgl Ⅱ enzyme digestion site, and trimethoprim resistance was obtained Plasmid(TPR-pK18mobSacB); The relA gene of Burkholderia pseudomallei HNBP001 was knocked out by homologous recombinant gene knockout method, and the relA mutant was obtained. The growth, motility and biofilm phenotypes of Burkholderia pseudomallei HNBP001 were compared before and after the deletion of relA gene. Results: The relA mutant was successfully constructed. The growth rate, motility and biofilm formation of ΔrelA decreased. Conclusion: The modified plasmid TPR- pK18mobSacB can improve the knockout efficiency of HNBP001 strain, which can be widely used in the gene knockout of Burkholderia pseudomallei, and provide convenience for the laboratory to study the gene function of Burkholderia pseudomallei at the molecular level; The mutant of the relA gene inhibits the growth, motility and biofilm formation of HNBP001.

    Screening microRNA expression profile in patients with acute T cell mediated rejection after kidney transplantation

    查看更多>>摘要:Objective: To explore the expression profile of miRNA in peripheral blood of patients with acute T cell mediated rejection after kidney transplantation. Method: 4 patients with acute T cell mediated rejection(TCMR) after kidney transplantation were collected as experimental group. Meanwhile,4 patients under stable condition after kidney transplantation were collected as control group. The elbow blood was taken from two groups. The next generation sequencing was used to study miRNA libraries. Then, they were analyzed of variance by R language software. Results: 14 miRNAs were differently expressed in the experimental group and control group, including 5 up-regulated miRNAs (P<0.05) and 9 down-regulated miRNAs (P<0.05). Conclusions: Peripheral blood miRNAs expression variations might be ideal biomarkers and reveal mechanisms for acute TCMR.

    查看更多>>摘要:Objective: We through the anatomy of cadavers to study the " Kambin's triangle " in the safe working area of lumbar intervertebral foramen and to provide anatomical reference for clinical lumbar fusion through Kambin's triangle approach. Methods: five complete cadaveric specimens were taken, the soft tissue of the lumbar back was removed, the transverse process, upper and lower articular processes and part of the vertebral lamina were bitten, the Kambin's triangle area of the lumbar spine was completely exposed, the bottom edge and height of the Kambin's triangle were measured, and the area of the Kambin's triangle was calculated; Using Kirschner wire, pull and fix the traveling nerve root to make the Kambin's triangle into a rectangle, measure the length of the bottom edge and height again, calculate the area, and compare the two groups of data. Results: the average height of the Kambin's triangle was 11.20mm ± 2.10mm, and the average height of the improved four corners was 11.19mm ± 1.93mm. The height of the improved four corners was slightly shorter than that of the Kambin's triangle. There was a significant correlation between the two, but the difference was not statistically significant. The average bottom of Kambin's triangle is 10.78mm ± 1.95mm, and the average bottom of improved four corners is 12.14mm ± 1.78mm. The length of the bottom edge of improved four corners is greater than that of Kambin's triangle. There is a significant correlation between them, and the difference is statistically significant; The average area of Kambin's triangle is 61.79mm2 ± 20.71mm2, and the area of improved four corners is 137.71mm2 ± 38.20mm2. The area of improved four corners is significantly larger than that of Kambin's triangle. There is a significant correlation between the two, and the difference is statistically significant. Conclusion: there is a narrow right angle triangle area surrounded by traveling nerve root, dural sac and superior endplate of lower vertebral body in the lumbar intervertebral foramen. If the traveling nerve root is pulled and fixed to turn the traditional Kambin's triangle into a quadrilateral, the bottom edge of the Kambin's triangle area can be significantly longer and the area can be significantly expanded, which can be operated more safely.

    Meta analysis of the effect of aerobic exercise on inflammatory factors and glucose and lipid metabolism in patients with type 2 diabetes mellitus

    查看更多>>摘要:Objective: To explore the effects of aerobic exercise on the levels of inflammatory factors in patients with type 2 diabetes. Methods: Randomized controlled studies (RCTs) on the effects of aerobic exercise on inflammatory factors in patients with type 2 diabetes were searched from electronic databases. Two researchers independently screened the literatures for data extraction and quality assessment. Meta-analysis was performed using RevMan 5.4 software. Results: A total of 17 literatures were included, including 1186 patients. The results of Meta-analysis showed that aerobic exercise significantly reduced the plasma levels of CRP, TNF-α, IL-6, and leptin in T2DM patients, but did not significantly reduce the plasma concentration of adiponectin. However, the left and right asymmetry of the "inverted funnel" plot indicated that there was a certain publication bias in the included studies. Conclusion: Existing evidence shows that aerobic exercise has a good effect on the improvement of chronic inflammatory state in patients with type 2 diabetes mellitus, suggesting that lifestyle changes can play a good role in adjunctive therapy.

    A mechanism to improve early diabetic nephropathy by modulating autophagy-related mechanisms with Yuye Decotion

    查看更多>>摘要:Objective: To investigate the molecular mechanisms of Yuye Decotion in the treatment of diabetic kidney disease using network pharmacology methods and molecular docking techniques. Methods: Obtain the transcriptome gene expression of diabetic nephropathy through GEO database, and extract genes related to autophagy. Screen the active ingredients and corresponding targets of Yuye Decoction through the TCMSP database, and map the drug prediction targets and disease targets to obtain the autophagy-related Yuye treatment targets for diabetic nephropathy point. Use String database combined with Cytoscape 3.7.2 software to construct the "drug-active ingredient-target" network and protein interaction network of Yuyetang for the treatment of diabetic nephropathy. The target point of liquid soup in the treatment of diabetic nephropathy was analyzed by GO biological process enrichment analysis and KEGG pathway enrichment analysis, and finally used Pymol and other software to analyze the core active components of Yuye Decotion and The core target protein undergoes molecular docking verification. Results: (i)100 eligible diabetic nephropathy and autophagy related genes were screened, and the potential targets of Yuye Decoction were 1,428. The acquired genes related to diabetic nephropathy and autophagy were mapped to potential targets of Yuye Decoction, and 22 therapeutic targets were obtained. GO biological process enrichment analysis and KEGG pathway enrichment analysis found that the pathways related to autophagy in the treatment of diabetic nephropathy by Yuye Decoction may include mTOR signaling pathway, phospholipase D signaling pathway, insulin resistance, EGFR tyrosine kinase inhibitor resistance, Apoptosis, PI3K /Akt signaling pathway, NF-κB signaling pathway, etc. (ii)The protein interaction network shows that VEGFA, ERBB2, GASP3, MAPK8, MYC, CDKN1A, EGFR, BCL2L1 may be the key targets of Yuye Decoction in the treatment of diabetic nephropathy. Molecular docking realizes the binding of 4 core active ingredients to 8 core target proteins. Conclusions: The research results show that Yuye Decoction treats diabetic nephropathy through multi-component, multi-target, and multi-pathway action, and provides new theoretical basis for the study of pharmacological effects and clinical application of Yuye Decoction in the treatment of diabetic nephropathy in autophagy-related aspects.

    查看更多>>摘要:Objective: To explore the mechanism and active components of Radix et Rhizoma Rhei in the treatment of Alzheimer's disease (AD) based on molecular docking. Methods: 22 major components of Radix et Rhizoma Rhei were screened from TCMSP and related literatures, which docked with the key targets of NLRP3/Caspase-1/GSDMD signaling pathway. NLRP3, Caspase-1, GSDMD inhibitors MCC950, ML132 and LDC7559 were used as positive control to analyze the docking results. Results: The docking results showed that the main components of Radix et Rhizoma Rhei had different degrees of binding with NLRP3, Caspase-1 and GSDMD targets, and the potential active components were mutanochrome and physciondiglucoside. Conclusion: Molecular docking predicts that the main components of Radix et Rhizoma Rhei may act on NLRP3/Caspase-1/GSDMD signaling pathway, and the active components may be mutanochrome and physciondiglucoside, which provides theoretical basis for revealing the anti-inflammatory mechanism and active components of Radix et Rhizoma Rhei in the treatment of AD.

    Based on the national patent database to explore the rule of traditional Chinese medicine in the treatment of hyperlipidemia

    查看更多>>摘要:Objective: To explore the medication law of traditional Chinese medicine compound patent for hyperlipidemia, and to provide reference for the research and development of traditional Chinese medicine new drugs for clinical treatment of hyperlipidemia. Methods: All compound TCM patents related to the treatment of hyperlipidemia were searched and extracted from the national patent database (http://epub.sipo.gov.cn) from the establishment of the database to 10st, March 2021. The data was established by the TCM Inheritance Support System (V2.5) and Microsoft Excel 2010 Softwore, and used to conduct TCM syndromes, frequency statistics, four nature and five flavors of traditional Chinese medicine, meridian attribution statistics.IBM SPSS statistics 20 software was used for drug clustering analysis, IBM SPSS modeler 18.2.2 software was used for association rules and web visualization analysis. Results: Totally 284 compound TCM patents were included, involving 577 kinds of Chinese medicine, and the most common syndrome was Yin deficiency of liver and kidney syndrome (74 times, accounting for 26.06%). The most ten common Chinese materiamedica was Hawthorn, Salvia miltiorrhiza, Alismaorientalis, Cassia seed, Polygonummultiflorum, Licorice, Poriacocos, chinese rhubarb, dried tangerine peel, Radix Paeoniae Alba. The top three appeared frequently in drug categories were tonifying deficiency medicine, activating blood-circulation and dissipating blood-stasis medicine, and heat-clearing medicine, respectively. The top two of its drug properties were warm and cold; the top three drug tastes were sweet, bitterness and pungent; the top two meridians were liver and spleen meridian, respectively. Seven drug groups were obtained by cluster analysis, and 16 were obtained by association rule analysis, including 5 drug pairs and 11 three drug groups. Conclusion: Through the data mining of the patent of traditional Chinese medicine compound prescription for regulating and treating hyperlipidemia, it is concluded that the fundamental pathogenesis of the disease is deficiency of viscera function and obstruction of phlegm and blood stasis. The clinical methods are tonifying the liver and kidney, activating blood circulation and removing stasis, resolving phlegm and removing dampness. Traditional Chinese medicine is mostly used for tonifying deficiency, activating blood circulation, removing stasis, resolving phlegm and removing dampness. This paper provides a reference for the research and development of traditional Chinese medicine for hyperlipidemia.

It is common in the elderly and has an increasing incidence.In 2021, the United States Association for the Study of Pain published new evidence-based clinical practice guidelines to provide more effective nonsurgical treatment of LSS-induced NC.Based on a thorough reading of the latest guidelines, combined with new clinical developments, and in collaboration with the 2011 North American Spine Society,NASS guidelines (hereinafter referred to as the 2011 Guidelines) were compared with the 2019 Danish Health Authority (DHA) Guidelines (hereinafter referred to as the 2019 Guidelines), and the clinical diagnosis and treatment of NC caused by LSS were reviewed and suggestions were put forward.
