    Effect of Shengsui Jiangu Capsule on bone conversion in rats with alcoholic osteoporosis

    查看更多>>摘要:Objective: To study the mechanism of Shengsui Jiangu Capsule promoting vitamin D3 regulating bone turnover in rats with alcoholic osteoporosis. Methods: 120 SD rats were randomly divided into four groups: model group (alcoholic osteoporosis model group), blank control group, western medicine control group (calcium carbonate tablet and affa D3 control group), Chinese medicine experimental group (Shengsui Jiangu capsule experimental group), 30 rats in each group. After 8, 12 and 16 weeks, the serum levels of 25 hydroxyvitamin D3 (25(OH)D3),1,25dihydroxyvitamin D3 (1,25(OH)2D3), cross-linked carboxy-terminal peptide of type 1 collagen (β-CTX) and N-terminal propeptide of type 1 collagen (P1NP) in rats and the density of the femur of rats were detected and compared. The correlation between vitamin D3 markers (25(OH)D3 and 1,25(OH)2D3) and bone turnover indices (P1NP and β-CTX) and bone mineral density was analyzed. Results: In the experimental group, bone mineral density (BMD) was significantly increased (P<0.01), bone repair and mineral stability and balance were maintained during bone turnover, the level of bone formation marker P1NP was significantly increased (P<0.01), and the level of bone resorption marker β-CTX was significantly decreased (P<0.01). Serum levels of 25(OH)D3 and 1,25(OH)2D3 were increased (P<0.01), and significantly higher than those in western medicine control group (P<0.05). Conclusion: Shengsui Jiangu capsule can effectively improve bone density in alcoholic osteoporosis (AOP) rats, and its mechanism may be related to promoting bone calcium absorption and regulating bone conversion.

    Exploring the mechanism of neuronal apoptosis and brain developmental damage in the hippocampus of hypoxicischemic neonatal rats based on BDNF/TrkB/CREB pathway

    查看更多>>摘要:Objective: Based on the BDNF/TrkB/CREB pathway, to explore the mechanism of neuronal apoptosis and brain developmental injury in the hippocampus of hypoxic- ischemic neonatal rats. Methods: Wistar young rats were ligated on one side of the common carotid artery and placed in an 8% oxygen and 92% nitrogen hypoxia box for 2 h to prepare hypoxic- ischemic brain injury models. Healthy rats were used as the control group. Control group and model group were selected, with 10 rats in each group, and the learning and memory ability was tested by Y-maze; 2,3,5-triphenyltetrazolium chloride (TTC) staining was used to detect brain tissue damage; Western blot was performed to determine the expression of brain-derived neurotrophic factor (BDNF), tyrosine protein kinase B (TrKB) and cAMP-response element binding protein (CREB) in hippocampal tissue. Another 15 mice in the control group and 60 mice in the model group were divided into negative control group (NC), BDNF overexpression group (LV-BDNF), TrkB overexpression group (LV-TrkB), and CREB overexpression group (LV-CREB), blank vector, BDNF, TrkB, CREB adenovirus overexpression vector was injected into the tail vein. Y-maze test for learning and memory ability; TTC staining method to detect brain tissue damage; neuronal apoptosis in the hippocampus were detected by terminal-deoxynucleoitidyl transferase mediated nick end labeling ; Western blot to detect the level of neuronal apoptosis in the hippocampus. Apoptosis-related protein B-cell lymphoma-2 (Bcl-2), BCL2associated X protein (Bcl-2 Assaciated X, Bax) and nuclear factor kappaB (NFκB) expression. Results: The learning and memory ability of the young mice in the model group was significantly reduced, the brain infarct volume was significantly increased, the expressions of BDNF and TrkB proteins in the hippocampus were significantly increased, and the expression of CREB proteins was significantly decreased; After overexpression of BDNF and TrkB CREB, in the LVBDNF, LVTrkB, and LVCREB group, the learning and memory ability of young mice were significantly improved, the brain infarct volume were significantly reduced, the hippocampal neuronal apoptosis were significantly reduced, The protein expression of Bax and NFκB were significantly decreased and the protein expression of Bcl2 were significantly enhanced. Conclusion: The expression of BDNF/TrkB/CREB is abnormal in HIBI model young mice. Overexpression of BDNF/TrkB/CREB can improve the learning and memory ability of young mice, repair brain tissue damage, and inhibit neuronal apoptosis. Therefore, the mechanism of HIBI may be related to BDNF/TrkB/CREB pathways.

    Simultaneous determination of ten components in Dianxiankang Capsules by HPLC QAMS

    查看更多>>摘要:Objective: To establish an high performance liquid chromatography-quantitative analysis of multi components by single marker(HPLC-QAMS) method for simultaneous determination of gastrodin, parishin E, parishin B, parishin, tenuifolin, onjisaponin B, methylophiopogonanone A, methylophiopogonanone B, β-asarone and α asarone in Dianxiankang Capsules. Methods: Waters Symmetry C18 column was used with acetonitrile-0.05 % phosphoric acid solution as mobile phase for gradient elution. Multiwavelength switching detection. The contents of gastrodin, parishin E, parishin B, parishin, onjisaponin B, methylophiopogonanone A, methylophiopogonanone B, β-asarone and α-asarone were calculated by relative correction factor. At the same time, the contents of 10 components in 12 batches of Dianxiankang Capsules were determined by external standard method (ESM). Results: An HPLC-QAMS method was established tenuifolin as the internal reference substance was established. The relative correction factors of gastrodin, parishin E, parishin B, parishin, onjisaponin B, methylophiopogonanone A, methylophiopogonanone B, β-asarone and α-asarone were 0.8238, 0.7239, 1.0229, 1.1881, 0.7272, 1.3108, 0.9314, 0.6549 and 1.0572, respectively. The relative correction factors had good repeatability and no significant difference with ESM(P>0.05). Conclusion: HPLC-QAMS can be used for simultaneous determination of multi-index components in Dianxiankang Capsules.

    Establishment and Validation of a nomogram for Predicting the Risk of Liver Fibrosis in Chronic HBV Infection

    查看更多>>摘要:Objective: To establish a non-invasive quantitative and visual predictive model for assessing the occurrence of significant fibrosis in chronic HBV infection, and to present nomogram to validate the efficacy. Methods: A total of 180 patients with chronic HBV infection that were admitted to the Department of Infectious Liver Diseases of the First Affiliated Hospital of Hainan Medical University from January 2019 to December 2021 with informed consent and underwent liver biopsy puncture were selected.131 patients and 49 patients were randomly divided into a model group and a validation group according to randomization. The patients were divided into non-significant fibrosis and significant fibrosis groups in the modeling group. To collect the clinic information, serological examination, liver elastography and liver histopathology results and to establish a rosette model to predict the risk of chronic HBV infection with significant fibrosis. Results: A total of 180 patients with chronic HBV infection were included, and 113 patients (62.7%) had significant fibrosis. In the modeling set,84 patients(64.1%) had significant fibrosis. In the modeled group, comparison of HBV DNA, PLT, ALT, AST, ALP, ALB, PAB, IL-6, HA, PⅢP, CIV, L.N and LSM for non-significant fibrosis and significant fibrosis showed statistically significant differences. The 氈2 values of the H-L goodness-of-fit test for the modelling and validation groups were 4.988 and 0.527, respectively, corresponding to P values of 0.08 and 0.77, suggesting that the nomogram has good predictive accuracy; the area under the ROC curve of the column line plot predicting the occurrence of significant fibrosis after HBV infection for the modelling and validation groups was 0.843[95%CI(0.775-0.910)] and 0.776[95%CI(0.714-0.838)], suggesting that the column line plot model has good discrimination. Conclusion: After stepwise regression analysis, it was established that ALB, HA, PⅢP, LSM and IL-6 were more closely associated with the occurrence of significant fibrosis after HBV infection, and a visualization of the occurrence of significant fibrosis column line graph model was established by comprehensive assessment, and validation was given that all were superior to the traditional models FIB-4 and APRI.

    Clinical observation of kidney tonifying and collateral tonifying acupuncture combined with thermosensitive moxibustion in the treatment of knee osteoarthritis and its effect on TRACP and CTX-I

    查看更多>>摘要:Objective: To observe the clinical effect of kidney-tonifying and colllateral-tonifying acupuncture combined with thermosensitive moxibustion in the treatment of knee osteoarthritis and cold congealing syndrome and the effect on Tartrate-resistant Acid Phosphatase (TRACP) and Ⅰ collagen C-terminal foreign body peptide (CTX-I). Methods:Totally 90 patients with knee osteoarthritis were divided into three groups: acupuncture group, thermosensitive moxibustion group and routine group with 30 cases in each group. In the conventional group, sodium hyaluronate was injected into the joint cavity and diclofenac sustained release tablets were taken orally. Thermosensitive moxibustion group was given thermosensitive moxibustion on the basis of conventional treatment. Acupuncture and moxibustion group in the heat sensitive moxibustion group treatment was given kidney tonifying acupuncture. VAS score, JKOM score and Ly-Sholm knee score were observed before and after treatment and followed up for 2 weeks. The levels of TRACP and CTX-I in serum before and after treatment. Results: Before treatment, there were statistically significant differences in VAS score, JKOM score and Ly-Sholm knee score among all groups (P>0.05). After treatment, the VAS score and JKOM score decreased, and the decrease degree was the greatest in the acupuncture group (P<0.05). Ly-Sholm knee score increased, and the change degree was the greatest in acupuncture group (P<0.05). Before treatment, the levels of TRACP and CTX-I in serum of all groups were significantly different (P>0.05). After treatment, serum TRACP and CTX-I levels were decreased, and the decrease degree was the greatest in acupuncture group (P<0.05). The total effective rates of the acupuncture group, the thermal sensitive moxibustion group and the conventional group were 96.7%, 80% and 66.7%, respectively, and the difference between each group was statistically significant (P<0.05). Conclusion: Kidney-tonifying and collature-tonifying acupuncture combined with thermosensitive moxibustion can significantly relieve knee pain and other symptoms of knee osteoarthritis in patients and cold congealing syndrome, which is worthy of clinical promotion.

    Real world analysis on renal impairment of elemene emulsion injection based on propensity score matching

    查看更多>>摘要:Objective: To explore the main factors of renal function injury caused by the combination of elemene emulsion injection in the real world. Methods: Based on the hospital information system medical database constructed by the Institute of Clinical Basic Medicine of Chinese Academy of Traditional Chinese Medicine, taking the patient information of elemene emulsion injection used in 21 top three hospitals in China as the research object, the nested case control study method was used to compare the combined medication of patients with renal function injury group and control group, The main factors that may lead to renal function injury were screened by MCP variable selection method, and the above main factors were further analyzed by classical logistic regression and propensity score logistic regression method. Results: The analysis found that the combined use of potassium chloride within 7 d before the use of elemene emulsion injection or the combined use of furosemide during the use of elemene emulsion injection may be the main factor leading to renal function injury. Conclusion: The combination of elemene emulsion injection and the above main factors that may lead to renal injury should be avoided. If it must be used, clinical monitoring should be strengthened, and abnormal reactions should be treated immediately. The results of this study have certain clinical significance, and the clinical mechanism can be further studied and explored in the future.

    Effectiveness and Safety of Chinese Medicine in the Treatment of PostTACE Syndrome of Primary Hepatocellular Carcinoma: A Systematic Review

    查看更多>>摘要:Objective: To systematically evaluate the effectiveness and safety of traditional Chinese medicine (TCM) in the treatment of posttransarterial chemoembolization (TACE) syndrome of primary liver cancer. Methods: CNKI, VIP database, Wanfang database, CBM, PubMed, MEDLINE, and Cochrane library were retrieved, and the RCTs of Chinese medicine in the treatment of primary liver cancer TACE postoperative syndrome were included from the establishment of the database to March 2022. The Cochrane bias risk tool was used to evaluate the quality of included articles. RevMan 5.4 software was used for metaanalysis of outcome indicators and GRADEprofiler 3.6 software was used to evaluate the quality of evidence. Results: A total of 3992 patients were included in 44 studies. Metaanalysis results showed that compared with conventional western medicine treatment, combined Chinese medicine treatment could reduce the incidence of fever [RR=1.45, 95%CI(1.09, 1.92)], nausea and vomiting [RR=1.47, 95%CI(1.10, 1.95)], liver pain [RR=1.21, 95%CI(1.02, 1.43)]; shorten the duration of fever [SMD=2.11. 95%CI(1.43, 2.79)], nausea and vomiting [SMD=1.67, 95%CI(1.22, 2.11)], hepatic pain [SMD=1.90, 95%CI(1.30, 2.51)], abdominal distension and pain [SMD=1.27, 95%CI(0.75, 1.79)], loose stool [SMD=2.30, 95%CI(1.24, 3.36)], constipation [SMD=2.18, 95%CI(1.52, 2.84)] after TACE,also it can improve patient quality of life [RR=0.73, 95%CI(0.65, 0.82)], clinical TCM syndrome [RR=0.73, 95%CI(0.65, 0.82)], shortterm efficacy [RR=0.75, 95%CI(0.66, 0.85)], oneyear survival rate [RR=0.87, 95%CI(0.78, 0.96)] and liver function and immune function. There was no significant difference in the incidence of abdominal distension and abdominal pain. Conclusion: Compared with only symptomatic western medicine after surgery, the combination of Chinese medicine has a significant effect and good safety in the treatment of postTACE syndrome.

    Clinicopathological significance and prognostic value of claudin10 expression in primary malignant tumors of digestive system: A metaanalysis

    查看更多>>摘要:Objective: To investigate the correlation between claudin10 expression and clinicopathological characteristics and prognosis of primary malignant tumors of digestive system. Methods: We searched the PubMed, Web of Science, Ovid MEDLINE, CNKI, CQVIP, and WanFang Data databases. The data between claudin10 expression and clinical characteristics as well as survival outcome were extracted. RevMan 5.3 and Stata 15.1 software were employed for metaanalysis. Results: A total of 6 studies containing 512 patients with primary malignant tumors of digestive system were analyzed. The analysis showed that high expression of claudin10 was more common in cancer tissue than in normal tissue[OR=3.22, 95%CI(2.26, 4.60), P<0.01], and was associated with increasing tumor size [OR=6.21, 95%CI(2.44, 15.83), P<0.01], high differentiation [OR=0.56, 95%CI(0.32, 0.99), P=0.05], positive lymph node metastasis [OR=3.41, 95%CI(1.09, 10.65), P=0.03]. The expression of claudin10 was not associated with sex[OR=1.62, 95%CI(0.91, 2.90), P=0.10],age[OR=1.72, 95%CI(0.97, 3.07), P=0.06], distant metastasis[OR=4.51, 95%CI(0.55, 36.70), P=0.16], or prognosis[HR=1.99, 95%CI(0.93, 4.23), P=0.08]. Conclusion: The expression of claudin10 is correlated with the clinicopathological features of primary malignant tumors of digestive system and is a potential tumor marker.

    Action mechanism of Yiqi Hexue Formula in the treatment of liver cirrhosis based on network pharmacology and experimental verification

    查看更多>>摘要:Objective: To study the mechanism of Yiqi Hexue Formula in the treatment of liver cirrhosis based on network pharmacology and experimental verification. Methods: Firstly, the effective components and action targets of various traditional Chinese medicines in Yiqi Hexue Formula were searched from TCMSP and BATMAN-TCM databases. The disease target genes were obtained by using GeneCard, PharmGkb, OMIM, DrugBank and TTD databases. The potential target genes of drugs and disease target genes were intersected, and the Venn diagram was drawn.The "active ingredient-core target" network was constructed by Cytoscape software, and the KEGG pathway of the intersection genes was analyzed by R software. Animal experiments were conducted to observe the effect of Yiqi Hexue Formula on histopathology of CCl4-induced liver cirrhosis model rats, determine the expression levels of JAK2 and STAT3 mRNA in liver tissue, and preliminarily explore the mechanism of Yiqi Hexue Formula in the treatment of liver cirrhosis. Results: A total of 43 active components and 648 targets of Yiqi Hexue Formula for the treatment of liver cirrhosis were screened through the database. KEGG pathway analysis showed that Yiqi Hexue Formula mainly affected PI3K/Akt, MAPK, IL-17 and JAK/STAT signaling pathway. Animal experimental research showed that compared with the model group, the degree of inflammation and fibrosis in liver tissue of rats treated with Yiqi Hexue Formula was significantly improved; The expression of JAK2 and STAT3 mRNA decreased significantly in the treatment group of Yiqi Hexue Formula (P<0.05).Conclusion: Yiqi Hexue Formula can treat liver cirrhosis through multiple components, multiple targets and multiple pathways. JAK2/STAT3 signaling pathway is involved in the formation of liver cirrhosis. Yiqi Hexue Formula can improve the pathological manifestations of cirrhotic rats. Its mechanism may be related to inhibiting the transmission of JAK2/STAT3 signaling pathway.

    Research progress on the mechanism of TCM treatment of depression based on NLRP3 pathway

    查看更多>>摘要:To review the research progress on the mechanism of traditional Chinese medicine (TCM) treatment of depression based on NLRP3 pathway in the past five years, in order to provide the basis for the clinical application of TCM treatment of depression. NLRP3, TCM and depression were used as key words to search PubMed and China Journal Fulltext Database (CNKI) and other databases for the relevant experimental literatures on antidepressant effects of TCM based on NLRP3 pathway in recent years. Chinese medicine can protect nerve cells by inhibiting the activation of NLRP3 and reduce the depression-like behavior in animal model. TCM can inhibit NLRP3 pathway and exert anti-depressive effect.
