    Insight into Metabolomic Profiling of Tinospora cordifolia:Recent Advances and Future Perspectives of Quality Control

    查看更多>>摘要:The objective of this study was to offer a review of previous works on Tinospora cordifolia(TC)metabolomics with a highlight on the use of metabolomics in comparative evaluations between varieties and selection of optimum cultivation,collection,and extraction procedures,providing insights into pharmaceutical and food product quality monitoring.Scholarly databases were searched to gather relevant information on TC's therapeutic usefulness,phytoconstituent studies,metabolomics,and applications.PubChem was used to learn the structures of several of the metabolites.The various medicinal properties of TC were extensively researched.The most essential portions of the plant in Ayurvedic medicines are the stems.Bioactive phytochemicals such as steroidal lactones,alkaloids,diterpenoids,and phenolic compounds are present in the roots and leaves.Metabolomic analysis revealed that,in addition to the season and geographical region of the sample collected,the metabolite profiles of TC also depend on the parts of the plant taken as a sample and also the plant's stage of growth.The amounts of secondary metabolites differed between morpho/chemotypes within the TC species.Despite the fact that research on TC began several decades ago,the diversity of phytoconstituents was not appreciated because of a lack of reliable techniques for phytochemical fingerprinting.The uncovering of several novel metabolites was aided by advances in chromatography combined with mass spectrometry.To produce a full metabolomic profile,researchers used mutually complementary techniques.Following that,data analysis and searches against spectrum databases allowed for signal annotation and interpretation of metabolites in large quantities without separating them separately.The current review includes a critical assessment of metabolomic data and an overview of the technique's various applications.It is vital to establish the metrics of quality control of herbal medicines in order to accomplish formulation purity assessments.Such information would assist us in determining the best geolocation for the plants and the best time to harvest them.Metabolomic investigation of TC-based herbal products is recommended for quality standards and the identification of novel bioactive components.

    查看更多>>摘要:The rich chemical constituents and diverse pharmacological properties of Ramulus Mori(RM)or the twig of Morus alba with evidence supported by clinical trials and patents are reviewed.Known as Sangzhi in Chinese,RM is widely used in traditional Chinese medicine to treat gout,arthritis,and rheumatism.Chemical constituents include flavonoids,benzofurans,alkaloids,stilbenes,chalcones,phenolic acids,and coumarins.Bioactivities of RM include antidiabetic,anti-obesity,anti-inflammatory,antityrosinase,neuroprotective,antioxidant,hepatoprotective,cytoprotective,renoprotective,antihyperuricemic,analgesic,antifungal,DNA protective,anticancer,gastroprotective,cardioprotective,anti-hair aging,anti-arthritic,and antiplatelet aggregation properties.The most active compounds from RM are mulberrin,oxyresveratrol,and mulberroside A.All four clinical trials on RM are focused on the treatment of diabetes.The patents entail using RM extracts as cosmetics for skin whitening and as agents for hypoglycemia.Topics for further research on RM are suggested.

    查看更多>>摘要:Background:Idiosyncratic drug-induced liver injury(IDILI)is a serious side effect of drugs,Epimedii Folium(EF)is unequivocally implicated in idiosyncratic liver injury onset,potentially due to its ability to perturb the NOD-like receptor family pyrin domain containing 3(NLRP3)inflammasome.Fructus Ligustri Lucidi(FLL),a frequently used medicinal combination with EF,has not yet been investigated for its ability to ameliorate EF-associated hepatotoxicity.Aims and Objectives:Study on the mechanism of compatibility of FLL to alleviate liver injury caused by EF.Materials and Methods:Western blot was used to determine the expression of related proteins,ELISA was used to detect the secretion of related inflammatory factors IL-1β,IL-18,IL-6 and TNF-α,liver injury indexes were detected and liver pathological tissue staining was used to evaluate the liver injury.Results:Our results demonstrated that EF exerted a particular augmenting effect on the stimulation of the NLRP3 inflammasome mediated by nigericin or ATP,whereas FLL suppressed the NLRP3 inflammasome stimulation.Furthermore,an equal EF to FLL ratio significantly reduced the stimulatory effects of EF.Moreover,EF has the potential to induce hepatic injury and augment pro-inflammatory cytokine synthesis in rats subjected to LPS.However,when combined with FLL,the detrimental effects of EF were mitigated.Conclusions:FLL possesses the capacity to attenuate EF-associated hepatotoxicity by suppressing EF-triggered NLRP3 inflammasome activation.Thus,FLL holds promise for improving the clinical safety profile of EF,shedding light on the potential of compatibility and detoxification theories in traditional Chinese medicine.

    查看更多>>摘要:Objective:The purpose of this research was to examine the potential anticancer properties of Xiao Tan San Jie Fang(XTSJF)and its potential mechanism of action against colorectal cancer.Materials and Methods:HCT116 cells were induced into HCT116 spheres in DMEM/F12 medium by treatment with epidermal growth factor+fibroblast growth factor+leukemia inhibitory factor+B27.The proliferation ability and stemness of HCT116 spheres was examined.Various concentrations of XTSJF were used to treat HCT116 spheres to observe the impact on proliferation,apoptosis,and expression of stem cell markers.Next,Wnt/β-catenin pathway-related factor proteins were detected.Results:The findings revealed that XTSJF suppressed cell growth and induced cell death in HCT116 cells in a dosage-dependent manner.Similarly,XTSJF promotes apoptosis,inhibits cell proliferation,prolongs survival,and maintains the expression of stem cells through the Wnt/-catenin/TCF4 axis.XTSJF also inhibits AKT activity and subsequently activates glycogen synthesis kinase-3β expression,inhibiting Wnt/beta-catenin pathway activity and downstream target gene transcript expression.The Wnt/β-catenin signaling pathway is inhibited by the XTSJF,leading to the suppression of colon cancer stem cell proliferation.Conclusion:Xiaotan Sanjie Prescription inhibited colon cancer stem cell growth through Wnt/β-catenin signaling pathway.

    Artificial Intelligence and Network Pharmacology Reveal the Medication Rules of Professor Wang Yu-Ying in the Treatment of Climacteric Syndrome

    查看更多>>摘要:Objective:The objective of this study was to explore the medication rules and academic ideas of Professor Wang Yu-Ying in the treatment of climacteric syndrome(CLS)and to predict new prescriptions.Materials and Methods:The characteristics of frequency,clustering,four properties,and five flavors were analyzed,and new prescriptions were predicted through an artificial intelligence(AI)-based method.The potential pathways of new prescriptions were explored through network pharmacology-based analysis.Results:The top 16 medicinals used by Professor Wang Yu-Ying in the treatment of CLS included Danggui,Longgu,Muli,Fuling,Chuanxiong,Gancao,Xiangfu,and Tusizi.The AI method was applied to predict the basic prescription for treating CLS:Danggui 15 g,Duanlonggu 30 g,Duanmuli 30 g,Fuling 28 g,Chuanxiong 10 g,Gancao 6 g,Xiangfu 12 g,Tusizi 14 g,etc.,Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses showed that the pathogenesis of CLS might be related to the estrogen pathway,involving typical steroid responses.Conclusions:This study summarized Professor Wang's medication experience in the treatment of CLS based on the data mining of clinical diagnoses and treatment cases.The AI method was used to predict the new prescription of CLS treatment,which was found to be reasonable by network pharmacology studies on its multi-target and multi-pathway mechanisms.

    Investigating the Potential Mechanism of Huazhi Rougan Granules Against Alcoholic Liver Disease by Network Pharmacology and Experiment

    查看更多>>摘要:Objective:Huazhi Rougan granules(HRGs)are a promising drug to improve alcoholic liver disease(ALD),but its mechanism remains unclear.Materials and Methods:Network pharmacology,molecular docking,and animal experiments were used to reveal the potential mechanism of HRG against ALD.Results:A total of 245 potential targets of HRG against ALD were obtained.Functional enrichment analysis suggests that HRG could synergistically regulate various biological pathways to exert therapeutic effects on ALD.Molecular docking showed that the key targets exhibited the good binding ability with the key compounds.The in vivo experiments showed that HRG can effectively alleviate pathological changes in liver tissue,improve blood lipid levels,antioxidant stress ability,and liver function,and reduce the release of inflammatory cytokines in the liver.Conclusions:This study reflects that HRG is an effective strategy for treating ALD,providing a basis for revealing the prevention and treatment mechanisms of ALD.

    查看更多>>摘要:Objective:This study aimed to explore andrographolide's mechanism of action and its protective effect on noise-induced hearing loss(NIHL).Materials and Methods:A mice animal model for NIHL was established through exposure to broadband noise at 120 dB sound pressure level for 4 h.Transcriptomics analysis and pharmacodynamic experiments were carried out.Results:Andrographolide enters the inner ear and effectively prevents hearing damage following noise exposure in the mice model for permanent hearing loss.Moreover,treatment with andrographolide inhibited the excessive activation of inflammatory factors in the cochleae of noise-exposed mice.Conclusion:Andrographolide might be a promising candidate for auditory protective drug investigation.

    查看更多>>摘要:Introduction:Dracaena cinnabari is considered a rich source of phytochemicals used widely in traditional medicine.In the present study,the effect of D.cinnabari hydraulic extract on the reproductive system of female rats was investigated.Methods:The samples were randomly divided into four groups(six samples in each group),including three treatment groups and one control group,and all samples were kept at the same conditions.Hydraulic extract of D.cinnabari and injected intraperitoneally daily for 10 days,while physiological serum was used for injection in to the control group.After 10 days of injection,estrogen and progesterone levels were measured by enzyme immunoassay technique.After dissection,the ovaries and uterine tissues were isolated for histological examination,and tissue changes were carefully examined.Results:The results revealed that the levels of estrogen and progesterone in experimental Groups 2 and 3 had a significant increase(P<0.001).Regarding tissue changes,a significant increase was observed in epithelial thickness(P<0.001),number of corpus luteum(P<0.01),and Graafian follicle(P<0.01)in doses of 100 and 150 mg/kg.Conclusions:Based on the results,it seems that D.cinnabari extract has an effect on the ovarian follicles.

    Comparative Evaluation of Antitumor Chinese Patent Medicine Injections Based on Evidence-Based Medicine and Markov Model

    查看更多>>摘要:Objective:The objective of this study was to explore the safety,efficacy,economy,and other aspects of frequently-used antitumor Chinese patent medicine injections through a comprehensive evaluation of evidence-based medical research to provide scientific information and decision-making basis for drug selection,rational drug use,public awareness of safe drug use,and medical insurance.Materials and Methods:The names of five frequently used antitumor Chinese patent medicine injections were used as search terms,including the compound Sophora flavescens injection,Kangai injection,Aidi injection,Kanglaite injection,and Brucea javanica oil emulsion injection.Relevant evidence-based medical research was extracted from the databases.A descriptive evaluation was conducted,and the Markov model was used for the cost-benefit analysis.Finally,the above injections are ranked by comprehensive scores using the multi-criteria decision analysis and Delphi method.Results:The five injections had their own advantages in enhancing the effectiveness of anti-tumor treatments,improving the quality of life for cancer patients,and reducing the incidence of adverse reactions.From the perspective of economy and other attributes,compound S.flavescens was superior to B.javanica oil,Aidi,Kangai,and Kanglaite.After a comprehensive evaluation of the above five injections,the compound S.flavescens injection received the highest score.Conclusions:Compound S.flavescens,Kangai,Aidi,B.javanica oil,and Kanglaite have their own advantages in effectiveness and no significant difference in safety.Compound S.flavescens injection has the highest comprehensive score.Clinical attention should be paid to monitoring the initial stage of medication and special groups.

    Uncovering Chemical Interactions between Danshen and Danggui Using Liquid Chromatography-Mass Spectrometry and Network Pharmacology-Based Research on Stroke

    查看更多>>摘要:Objective:The objective of this study was to decipher chemical interactions between Danshen and Danggui using liquid chromatography-mass spectrometry(LC-MS)and explore the mechanisms of Danshen-Danggui against stroke using network pharmacology and molecular docking.Materials and Methods:First,the chemical compounds of Danshen-Danggui were profiled using ultra-high-performance liquid chromatography(HPLC)-quadrupole time-of-flight MS.Accurately characterized compounds in various proportions of Danshen-Danggui were quantified using HPLC combined with triple quadrupole electrospray tandem MS.Network pharmacology was used to uncover the essential mechanisms of action of Danshen-Danggui against stroke.Discovery Studio Software was used for the molecular docking verification of key active chemicals and stroke-related targets.Results:A total of 53 compounds were characterized,and 22 accurately identified constituents(10 phenolic acids,8 phthalides,and 4 tanshinones)were quantified in 15 proportions of Danshen-Danggui.The quantification results showed that Danggui significantly increased the dissolution of most phenolic acids(compounds from Danshen),whereas Danshen promoted the dissolution of most phthalides(compounds from Danggui).Overall,the combination of Danshen and Danggui at a 1:1 ratio resulted in the maximum total dissolution rate.Further network pharmacology and molecular docking results indicated that Danshen-Danggui exerted anti-stroke effects mainly by regulating inflammation-related(tumor necrosis factor,hypoxia-inducible factor,and toll-like receptor)signaling pathways,which ranked among the top three pathways based on Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis.Conclusion:The chemical compounds in Danshen-Danggui could interact with each other to increase the dissolution of the most active compounds,which could provide a solid basis for uncovering the compatibility mechanisms of Danshen-Danggui and Danshen-Danggui-based formulae.
