    Microbiomics and metabolomics insights into the microbial regulation on the formation of flavor components in the traditional fermentation process of Chinese Hongqu aged vinegar

    查看更多>>摘要:This study aimed to investigate microbial succession and metabolic dynamics during the traditional fermentation of Hongqu aged vinegar,and explore the core functional microbes closely related to the formation of flavor components.Microbiome analysis demonstrated that Lactobacillus,Acetobacter,Bacillus,Enterobacter,Lactococcus,Leuconostoc and Weissella were the predominant bacterial genera,while Aspergillus piperis,Aspergillus oryzae,Monascus purpureus,Candida athensensis,C.xylopsoci,Penicillium ochrosalmoneum and Simplicillium aogashimaense were the predominant fungal species.Correlation analysis revealed that Acetobacter was positively correlated with the production of tetramethylpyrazine,acetoin and acetic acid,Lactococcus showed positive correlation with the production of 2-nonanone,2-heptanone,ethyl caprylate,ethyl caprate,1-hexanol,1-octanol and 1-octen-3-ol,C.xylopsoci and C.rugosa were positively associated with the production of diethyl malonate,2,3-butanediyl diacetate,acetoin,benzaldehyde and tetramethylpyrazine.Correspondingly,non-volatile metabolites were also detected through ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry.A variety of amino acids and functional dipeptides were identified during the traditional brewing of Hongqu aged vinegar.Correlation analysis revealed that Lactobacillus was significantly associated with DL-lactate,indolelactic acid,D-(+)-3-phenyllactic acid,pimelic acid,pregabalin and 3-aminobutanoic acid.This study is useful for understanding flavor formation mechanism and developing effective strategies for the suitable strains selection to improve the flavor quality of Hongqu aged vinegar.

    High β-sitosterol-D-glucoside content in sweet potato varieties and its anti-breast cancer potential through multiple metastasis-associated signals

    查看更多>>摘要:β-Sitosterol-D-glucoside(β-SDG)is a phytosterol compound whose antitumor activity has been confirmed by previous studies.However,its suppression on breast cancer remains unclear.To that purpose,we isolated β-SDG from sweet potato and investigated the breast-cancer-inhibiting mechanism using proteomic analysis.The sweet potato species S6 with high β-SDG content were chosen form 36 species and β-SDG was isolated by HPLC.Afterwards,an in situ animal model of breast cancer was established,and β-SDG significantly reduced the tumor volume of MCF-7 xenograft mice.Proteomic analysis of tumor tissues revealed that 127 of these proteins were upregulated and 80 were downregulated.Gene ontology and network analysis showed that regulatory proteins were mainly associated with epithelial-mesenchymal transition(EMT),myogenesis,cholesterol homeostasis,oxidative phosphorylation and reactive oxygen pathways,while Vimentin,NDUF,VDAC1,PPP2CA and SNx9 were the most significant 5 node degree genes.Meanwhile,in vitro and in vivo results showed that the protein expression of PPP2CA and Vimentin,which are markers of EMT,were involved in breast cancer cell metastasis and could be reversed by β-SDG.This work highlights β-SDG as a bioactive compound in sweet potato and the potential therapeutic effect of β-SDG for the treatment of breast cancer by inhibiting metastasis.

    Effects of salt and rice flour concentration on microbial diversity and the quality of sour meat,a Chinese traditional meat

    查看更多>>摘要:This study investigated the effects of salt(3%and 6%,m/m)and rice flour(10%and 20%,m/m)addition in sour meat,a traditional Chinese fermented meat.It was found that salt has greater effect than rice flour addition in spontaneous fermentation.Low-salt groups had lower pH and higher titratable total acid.In the low-salt groups,the dominant genera were Lactobacillus and Lactococcus,whereas Staphylococcus,Weissella,and Tetragenococcus were dominant in the high-salt groups.Higher total free amino acids and essential amino acids,organic acids,hexanoic acid ethyl ester and octanoic acid ethyl ester were found in the low-salt groups.The RDA analysis revealed that Lactococcus was closely related to product quality,with the S3F10(3%salt and 10%rice f lour)group outperforming the others in the sensory evaluation.Therefore,3%salt and 10%rice flour were considered more appropriate for the production of healthy and tasty fermented sour meats.

    查看更多>>摘要:Lactobacillus rhamnosus(Rh)and Lactobacillus reuteri(Re)are well-known probiotic species in inflammatory bowel disease(IBD)research.The variations between these species'efficacy against colitis,and their model of action in this regard,are intriguing and enable treatment to be individually tailored to patients.In this study,four strains each of Rh and Re were isolated from fecal samples and their draft genomes were sequenced.The anti-colitis activities of both strains involved various aspects of intestinal immune,physical,chemical,and biological barrier function.Strikingly,the tested strains exhibited considerable interspecies and intraspecies specificity in colitis amelioration.Rh strains significantly outperformed Re strains in terms of short-chain fatty acid synthesis.Nevertheless,Re strains were more effective than Rh strains in inhibiting production of inflammatory factors;promoting production of intestinal mucus,antimicrobial peptides,and tight junction proteins;and supporting the stem cell compartment.This accounts for the anti-colitis outcomes of Re strains being superior to those of Rh strains.In addition,the effective Rh and Re strains were found to express high concentrations of specific carbohydrate metabolism-and prophage-related genes,respectively.Taken together,the results of this study could assist researchers in developing effective therapies for IBD.

    查看更多>>摘要:Fufang E'jiao Jiang(FEJ)as a healthy food consisting of medicine food homology materials approved by China's Ministry of Health has been extensively applied to replenish qi and nourish blood,and it has a positive impact on women's health.To find out the material basis and mechanism of FEJ,a systematic"compound-effect-target"analysis including chemical composition resolution,zebrafish,network pharmacology,molecular docking,transcriptome,and bibliometric analysis was adopted.124 chemical components including ginsenosides,and phenylethanoid glycosides in FEJ were discovered,and effects of FEJ on promoting the generation of immune cells,erythropoiesis and angiogenesis in zebrafish were exhibited.Based on network pharmacology,molecular docking and in vivo activity assay,6 compounds including jionoside A1,isoacteoside,echinacoside,acteoside,lobetyolin,and rehmannioside D were identified as active components of FEJ.Transcriptome data showed that several pathways such as complement and coagulation cascades,ECM-receptor interaction,and PI3K-Akt signaling pathway were associated with proangiogenic effect of FEJ.19 common targets were obtained through combined analysis of network pharmacology and transcriptomics,and 5 targets of them were verified by PCR.The bibliometric analysis of these common targets revealed that FEJ was related to energy metabolism,pathway in cancer,etc.,which was consistent with the results of network pharmacology and transcriptome.The studies suggested that FEJ could replenish qi and nourish blood through multi-compound and multi-targets.

    Lactobacillus from fermented bamboo shoots prevents inflammation in DSS-induced colitis mice via modulating gut microbiome and serum metabolites

    查看更多>>摘要:Fermented bamboo shoots(FBS)is a region-specific food widely consumed in Southwestern China,with Lactobacillus as the predominant fermenting bacteria.However,the probiotic potential of Lactobacillus derived from FBS reminds largely unexplored,especially for diseases with a low prevalence in areas consuming FBS,namely,inflammatory bowel disease.In this study,Lactiplantibacillus pentosus YQ001 and Lentilactobacillus senioris YQ005 were screening by in vitro probiotic tests to further investigate the probiotic-like bioactivity in dextran sulfate sodium(DSS)-induced ulcerative colitis(UC)mouse.They exhibited more positive probiotic effects than Lactobacillus rhamnosus GG in preventing intestinal inflammatory response.The results revealed that both strains improved the abundance of deficient intestinal microbiota in UC mice,including Muribaculaceae and Akkermansia.In the serum metabolome,they modulated the DSS-disturbed levels of metabolites,with significant increment of cinnamic acid.Meanwhile,they reduced the expression levels of interleukin-1β(IL-1β),interleukin-6(IL-6)inflammatory factors and increased zonula occludens-1(ZO-1),Occludin,and cathelicidin-related antimicrobial peptide(CRAMP)in the colon.Consequently,these results demonstrated that Lactobacillus spp.isolates derived from FBS showed promising probiotic activity based on the gut microbiome homeostasis modulation,anti-inflammation and intestinal barrier protection in UC mice.

    查看更多>>摘要:Atherosclerosis(AS)is the main pathological basis of cardiovascular diseases.Hence,the prevention and treatment strategies of AS have attracted great research attention.As a potential probiotic,Pararabacteroides distasonis has a positive regulatory effect on lipid metabolism and bile acids(BAs)profile.Oligomeric procyanidins have been confirmed to be conducive to the prevention and treatment of AS,whose anti-atherosclerotic effect may be associated with the promotion of gut probiotics.However,it remains unclear whether and how oligomeric procyanidins and P.distasonis combined(PPC)treatment can effectively alleviate high-fat diet(HFD)-induced AS.In this study,PPC treatment was found to significantly decrease atherosclerotic lesion,as well as alleviate the lipid metabolism disorder,inflammation and oxidative stress injury in ApoE-/-mice.Surprisingly,targeted metabolomics demonstrated that PPC intervention altered the BA profile in mice by regulating the ratio of secondary BAs to primary BAs,and increased fecal BAs excretion.Further,quantitative polymerase chain reaction(qPCR)analysis showed that PPC intervention facilitated reverse cholesterol transport by upregulating Srb1 expression;In addition,PPC intervention promoted BA synthesis from cholesterol in liver by upregulating Cyp7a1 expression via suppression of the farnesoid X receptor(FXR)pathway,thus exhibiting a significant serum cholesterol-lowering effect.In summary,PPC attenuated HFD-induced AS in ApoE-/-mice,which provides new insights into the design of novel and efficient anti-atherosclerotic strategies to prevent AS based on probiotics and prebiotics.

    Longitudinal investigation of mineral composition in human milk and its correlation with infant anthropometric outcomes among Tibetan mother-infant dyads during the first 6 months postpartum

    查看更多>>摘要:Nutrients in human milk,including minerals,relate growth and development of breast-fed infants.Tibetan mother-infant dyads possess unique characteristics on early nutrition due to their featured long-lasting life-style.This study longitudinally investigated the relationship between the mineral composition in human milk and the Z-scores of infants among Tibetan mother-infant dyads during their first 6 months postpartum through a prospective cohort study.The results show that the minerals of Na,Mg,K,Ca,Cu,Zn,and Se were of higher levels in colostrum than other lactation stages.Several minerals were below the recommended values for infants according to Chinese dietary guidelines.Besides,a large proportion of infant Z-scores were below-2 as lactation period continued.Multivariate statistical analysis revealed that classifications and correlations in varying degrees were observed between minerals in human milk and infant Z-scores.These findings will be advantageous for research upon Chinese early nutrition and progress of tailor-made infant formula.

    查看更多>>摘要:Commercial sterility does not guarantee the sustained stability of ultrahigh temperature(UHT)milk over 6 months shelf life.We explore the microbiota presented in normal(SZ)and quality deteriorated UHT milk(QY and WY)products from the same brand.Based on high-throughput sequencing research results,11 phyla and 54 genera were identified as dominant microbiota.Pseudomonas,Streptococcus,and Acinetobacter as core functional microbiota significantly influenced the UHT milk quality properties.Moreover,principal component analysis(PCA)and multivariate analyses were used to examine the quality characteristics,including 11 physicochemical parameters,10 fatty acids,and 2 enzyme activities,in normal and quality deteriorated UHT milk.We found that the abundance of Pseudomonas increased in quality deteriorated milk(WY)and showed a significant positive correlation with heat-resistant protease content.Acinetobacter in quality deteriorated milk(QY)also considerably contributed to the content of heat-resistant lipase,which resulted in spoilage deterioration of UHT milk.

    Strain-specific effect of Streptococcus thermophilus consumption on host physiology

    查看更多>>摘要:Streptococcus thermophilus is one of the most prevalent species in stool samples of westernized populations due to continuous exposure to fermented dairy products.However,few studies have explored the effect on host physiology by multiple S.thermophilus strains and considered the inter-strain differences in regulating host.In the present study,we investigated how four S.thermophilus strains influenced the gut microbiota,mucin changes,and host metabolism after 28 days of intervention in conventional mice.The results indicated that the consumption of S.thermophilus affected the host with strain specificity.Among four S.thermophilus strains,DYNDL13-4 and DQHXNQ38M61,especially DQHXNQ38M61,had more effect on host physiology by modulating gut microbiota and host metabolism than LMD9 and 4M6.Ingestion of strains DYNDL13-4 and DQHXNQ38M61 resulted in more remarkable changes in amino acid metabolism and lipid metabolism than that of strains LMD9 and 4M6,which may be related to the elevation of intestinal Bifidobacterium by DYNDL13-4 and DQHXNQ38M61.The enriched Coriobacteriaceae UCG-002,Rikenellaceae RC9 gut group,and Lactobacillus only in the DQHXNQ38M61 group,had a close relationship with the prominent effect of DQHXNQ38M61 on regulating amino acid and lipid metabolism.In addition,DQHXNQ38M61 had a strong influence on degrading colonic mucin fucose by decreased α-fucosidase activity in feces,and improving mucin sulfation by upregulated Gal3ST2 expression.Comparative genomic analysis revealed that the four S.thermophilus strains belonged to different branches in the phylogenetic tree,and DYNDL13-4 and DQHXNQ38M61 had more genes involved in carbohydrate metabolism,amino acid metabolism,membrane transport,and signal transduction,which may confer the capacity of nutrient utilization and gastrointestinal adaptation of the strains and be associated with their strong regulation in host.Our study provides valuable information for understanding the regulation of host metabolism after consuming different S.thermophilus strains and could facilitate potential personalized applications of S.thermophilus based on strain varieties.
